Objective: To observed the effect of evening primrose oil treatment on rabbit acne model and explore its regulation effect on p38 MAPK and NF-κB signaling pathway. Methods: Coal tar was coated on rabbit ear to establish the rabbit ear acne model, 1 time a day, for 15 days.Acne models were randomly divided into the control group, the positive control group, evening primrose oil groups (1. 77 g/kg, 1. 18 g/kg, 0.59 g/kg), tanshinone group (0. 21 g/kg). The treatment effect was determined by HE staining, and the immunohistochemical method was used to detect expressions of NF-κB and p38 MAPK protein. ELISA method was used to detect changes of IL-1αand IL-6 in serum. Results:Compared with the control group, contents of IL-1α and IL-6 in the model group significantly increased, and the difference was statistically significant (P <0. 01). Compared with the control group, contents of IL-1α and IL-6 in each drug group significantly decreased, and differences were statistically significant (P <0. 01). Compared with the control group, the average optical density of p38 MAPK and NF-κB protein in the model group significantly increased, and the difference was statistically significant (P <0. 01). Compared with the model group, average optical densities of p38 MAPK and NF-κB protein in each drug group significantly decreased (P <0. 01). Average optical densities of p38 MAPK and NF-κB protein in 1. 77 g/kg evening primrose oil significantly decreased, while contents of IL-1α and IL-6 significantly decreased. Conclusion: Evening primrose oil can reduce the formation of hair follicles keratin plug in the rabbit ear, lighten the cuticles, inhibit ductal epithelial hyperplasia of sebaceous gland, reduce inflammatory cells infiltration. The mechanism is to control the serum IL-1α and IL-6 through p38 MAPK and NF-κB signaling pathway.%目的:观察月见草油治疗兔耳痤疮模型的效果并探讨其对p38 MAPK、NF-κB信号通路的调节作用.方法:在兔耳内涂抹煤焦油使之形成痤疮, 每天1次, 持续15日, 造模成功后随机分为空白组, 模型组 (阳性对照组), 月见草油1.77g/kg、1.18g/kg、0.59g/kg组及丹参酮0.21g/kg组.通过HE染色显微镜下判定其治疗效果;NF-κB、p38 MAPK蛋白的表达用免疫组化检测;IL-1α、IL-6在血清中的含量变化通过ELISA测算.结果:与空白组比较, 模型对照组IL-1α、IL-6含量显著升高;与模型组比较, 各给药组IL-1α、IL-6含量均明显下降.与空白组比较, 模型组p38 MAPK、NF-κB蛋白平均光密度值增高;与模型组比较, 各给药组p38MAPK、NF-κB蛋白平均光密度值降低明显.结论:月见草油治疗痤疮的疗效确切, 能减少兔耳毛囊脂质栓塞的同时减轻其扩张并抑制皮脂腺导管上皮增生, 减少炎症细胞浸润.其机制为通过p38 MAPK、NF-κB信号通路来调控血清IL-1α、IL-6.
