    • 摘要: 目的 超高效液相色谱-三重四级杆质谱串联(UPLC-TQD-MS)法探讨关木通配伍干姜减毒存效机制.方法 关木通配伍干姜的分析采用ACQUITYUPLC BEH@C8色谱柱(2.1 mm×50mm,1.7 μm);流动相1mmol/L乙酸铵(含0.196乙酸)-甲醇;体积流量0.2mL/min;柱温30°C;采用电喷雾离子源正离子模式(ESI+)和多反映监测模式(NRM)进行质谱分析.结果 马兜铃酸A在0.809 ~51.750μg(r=0.999 5)的范围内线性关系良好,平均加样回收率为98.18%,RSD为1.4%.关木通-干姜(1∶2)配伍使用时关木通中马兜铃酸A显著降低,降低率达到87.79%,对大鼠的利尿作用没有变化,对大鼠急性肾功能损伤程度明显低于关木通生品组.结论 该方法准确稳定,重复性好,可用于关木通配伍干姜的质量控制.

    • 摘要: Direct spray mass spectrometry was used to simply and rapidly differentiate Mutong of Aristolochiaceae from other two kinds of Mutong medicinal materials (Lardizabalaceae and Ranunculacea) by analyzing the chemical profile of Mutong of Aristolochiaceae.A novel method for determination of magnoflorine content in Mutong of Aristolochiaceae was established.The results showed that Mutong of Aristolochiaceae could be identified according to the symbolic component, magnoflorine.Under positive ion mode, semi-quantitative result based on the signal strength ratio of magnoflorine and nuciferin was obtained by choosing nuciferin as an internal standard.The method showed good linear coefficient in the concentration range of 0.50-20.00 mg/L of magnoflorine.The limit of detection was 0.1 mg/L.The method was simple and fast, and could be used for direct and rapid in-situ analysis and identification of Mutong of Aristolochiaceae from other closely related Mutong herbal species without sample pre-treatment.The results were important for the quality control of Mutong herbal medicine.%采用直喷离子化质谱技术,通过对关木通的敞开式质谱轮廓分析,对关木通和其它两种木通(木通、川木通)进行了快速鉴别,建立了关木通中木兰花碱的快速测定方法.结果表明,可以利用标志性成分木兰花碱对马兜铃科关木通进行鉴别;同时,在正离子模式下,以荷叶碱为内标,木兰花碱与内标的信号强度比与木兰花碱标准溶液的浓度,在0.50~20.00 mg/L范围内线性关系良好,相关系数为0.9989,检出限为0.1 mg/L. 本方法简单、快速、无需样品前处理,可用于有毒药材关木通的直接、原位、快速分析和鉴定.本研究结果对木通中药材的质量控制具有重要参考价值.

    • 摘要: 目的:掌握木通、川木通、关木通的鉴别方法,确保购迸合格的原料药材,提升中成药的安全性和有效性.方法:从性状鉴别、显微鉴别和薄层鉴别等方面对木通、川木通、关木通迸行鉴别和对比.结果:外观性状:木通切面皮部较厚,黄棕色,髓部较小或有时中空,黄白色或黄棕色,周边灰棕色至灰褐色;川木通残存皮部棕黄色,髓部较小,类白色,偶有空腔,周边浅黄色;关木通皮部薄,髓部不明显,周边灰黄色.显微鉴别:木通含晶石细胞方形或长方形,胞腔内含1至数个棱晶;川木通石细胞类长方形、梭形或类三角形;关木通石细胞少见,含淀粉粒和草酸钙簇晶.薄层色谱法也能很好地将木通与川木通、关木通迸行区分.结论:性状鉴别,显微鉴别和薄层鉴别能很好地将木通、川木通和关木通区分,从而保证临床用药的安全和有效.

    • 摘要: Objective To investigate the changes of early biomarkers, such as neutrophil gelatinase-associated lipocalin( NGAL) , Cystatin C in blood and urine, as well as changes of total protein( TP) , albumin( ALB) in urine, in a rat model of acute kidney injury induced by Caulis Aristolochiae Manshuriensis(CAM).Methods Twenty-four male SD rats were randomly divided into 3 groups:the control group, the aristolochic acid-Ⅰ ( AAⅠ) group and the CAM group.Rats in the 3 groups received treatment with water, AAⅠat 36 mg/kg or CAM at 60 g/kg through gastric infusion for 3 days followed by a two-day break.Venous blood samples and urine samples were collected 1 day before treatments, 6 h and 12 h after each treatment.Blood, 24 h urine and kidney tissues were collected on the 5th day.At different time points, SCr, BUN, TP, GGT and ALB in urine or serum were measured.Secretion of NGAL and Cystatin C in urine or serum were determined by ELISA.Samples of kidney tissue were observed after HE staining.Results In the AAⅠ and CAM groups, urinary NGAL, TP, ALB and Cystatin C showed a significant increase and an upward trend with time ( P<0.05).Conclusion NGAL, TP, ALB and Cystatin C may be used as biomarkers for early diagnosis of kidney injury in-duced by CAM.%目的:应用关木通水提液诱导大鼠急性肾损伤模型,观察早期生物标志物胱抑素C( Cystatin C)、中性粒细胞明胶酶相关脂质运载蛋白( NGAL)在血和尿中的动态变化以及总蛋白( TP)、白蛋白( ALB)在尿液中的变化。方法将24只SD大鼠随机分为对照组、马兜铃酸-Ⅰ( AAⅠ)组和关木通组,每组8只,分别用蒸馏水、AAⅠ混悬液和关木通水提液灌胃给药,连续给药3 d,停药2 d,实验周期5 d。在实验前1 d,每次给药后6、12 h自大鼠内眦静脉取血,用代谢笼留取相应时间点尿液,第5天收集24 h尿液、血液和肾组织。用ELISA法检测早期生物标志物Cystatin C和NGAL,酶法检测肌酐、尿素氮、TP、谷氨酰转肽酶和ALB,HE染色观察大鼠肾脏形态学变化。结果关木通组大鼠在给药第1天12 h血清和尿液NGAL,第2天12 h尿液Cystatin C、尿TP、尿ALB,第3天12 h血肌酐和尿素变化均较对照组升高( P<0.05)。 AAⅠ组大鼠在给药第1天12 h尿NGAL,第2天12 h尿ALB,第3天12 h血清Cystatin C、尿液TP均较对照组升高( P<0.05)。结论检测血和尿中早期生物标志物水平有助于及时发现肾损伤。

    • 摘要: 目的:为了确定中药材关木通的最佳减毒方法,探讨不同炮制方法对中药材关木通中马兜铃酸的脱除效果。方法对中药材关木通进行姜炙、醋炙、蜜炙、甘草炙、碱制、炒炙、酒炙以及碱蜜合制、碱酒合制、碱姜合制,采用反相高效液相色谱法测定马兜铃酸的含量,计算马兜铃酸的脱除率。结果醋炙关木通药材中马兜铃酸的含量为(0.101000±0.001700)%,醋炙品水浸出物与醇浸出物的含量相当。结论醋炙制法为最佳方法。%Objective To explore removal effect of aristolochic acid with different processing methods on Aristolochia Manshuriensis in order to determine the best attenuated method of Aristolochia Manshuriensis. Methods Main ginger, vinegar main, honey main, licorice broiled, alkali system, fried grilled, broiled, alkali and honey also, alkali and liquor also, alkali and ginger also were applied in Aristolochia Manshuriensis. Content of aristolochic acid in Aristolochia Man-shuriensis was detected by RP-HPLC, and removal rate of aristolochic acid was calculated. Results Content of aris-tolochic acid in Aristolochia Manshuriensis with vinegar main was (0.101 000±0.001 700)%, the content of water extract and alcohol extract in vinegar main was quite. Conclusion This vinegar processing is the best way to take effect.

    • 摘要: 目的:通过反相高效液相色谱法观察比较关木通配伍苦寒药黄连后马兜铃酸A的变化趋势,探讨中药配伍对马兜铃酸A的影响.方法:色谱柱为Diamonsil C18(4.6 mm×250 mm,5μm),以乙腈:(水+冰醋酸)(体积比51∶49)为流动相,检测波长250 nm.结果:关木通与黄连配伍后,各配伍组马兜铃酸A含量有一定程度的降低,其中以关木通与黄连配伍比例为(1∶1.5)时,AAI含量下降最为明显,达72%.结论:“寒寒相配”的中药配伍形式有助于减低关木通中马兜铃酸A的含量,中药配伍减毒理论有一定的科学依据.
