    • 摘要: 目的 探讨七氟醚(sevoflurane)对心脏骤停(cardiac arrest,CA)大鼠的脑保护机制.方法 建立室颤引起的大鼠心脏骤停模型,40只雄性Wistar大鼠随机分为假手术组(Sham)、七氟醚组(Sevo组)和对照组(Control).自主循环恢复(restoration of spontaneous circulation, ROSC)后24 h,用Western blot方法检测凋亡相关蛋白Bax、Bak、Bcl-2、细胞色素c、裂解caspase 9及裂解caspase 3的表达;ROSC 24 h分光光度计法测线粒体通透转换孔(mitochondrial permeable transfer hole,MPTP)开放程度,JC-1荧光探针测线粒体膜电位(mitochondrial membrane potential,MMP);ROSC 72 h以TUNEL法检测海马CA1区神经元凋亡.结果 Sevo组Bax、Bak、裂解caspase 9和裂解caspase 3和胞质中细胞色素c的含量低于对照组(均P<0.05),Bcl-2表达高于对照组(P<0.05);Sevo组MPTP开放程度低于对照组,MMP高于对照组(均P<0.05);Sevo组神经元凋亡指数低于对照组(P<0.05).结论 七氟醚可能通过下调Bax和Bak,上调Bcl-2的表达,减少细胞凋亡并抑制MPTP开放程度,从而发挥脑保护作用.%Objective To investigate the neuroprotective mechanism of sevoflurane in rats resuscitated from cardiac arrest (CA).Methods A ventricular fibrillation-induced CA model was established.Forty Wistar rats were randomly divided into the sham group,sevoflurane group and control group.Apoptosis-related proteins were measured by Western blot at 24 h after restoration of spontaneous circulation (ROSC).The status of mitochondrial permeability transition pore (MPTP) were measured using a spectrophotometer,and the mitochondrial membrane potential (MMP) were measured with JC-1 fluorescent probe.At 72 h after ROSC,the apoptotic index of neurons in hippocampal CA1 region was counted by TUNEL staining.Results The protein expression of Bax,Bak,cleaved-caspase 9,cleavedcaspase 3 and cytosolic cytochrome c were lower in the sevoflurane group (all P<0.05),the protein expression of Bcl-2 was higher in the sevoflurane group compared with the control group (P<0.05).The sevoflurane group had a less opening status of MPTP and a higher MMP compared with the control group (all P<0.05).The sevoflurane group had less apoptotic neurons compared with the control group (P<0.05).Conclusion By up-regulating the expression of Bcl-2,down-regulating Bax and Bak,sevoflurane could reduce the apoptosis of neurons and decrease the opening of MPTP,eventually reduce cerebral injuries.

    • 摘要: 目的 探讨单侧咀嚼力降低对大鼠颞下颌关节软骨中caspase-12和Bak表达的影响.方法 选取96只3周龄雄性SD大鼠,随机分为实验组及对照组各4组,每组12只.实验组大鼠行左侧所有磨牙拔除术,对照组大鼠不做任何处理,分别于拔牙后1d,7d,14d,28d处死.HE染色观察软骨的形态学变化,RT-PCR方法检测caspase-12和Bak在关节软骨中的表达.结果 实验组大鼠髁突软骨细胞随时间延长,逐渐出现细胞分层模糊,纤维断裂,软骨细胞空泡性变等退行性变化.拔牙后,实验组大鼠髁突软骨细胞的caspase-12和Bak的含量均高于对照组,差异有统计学意义(P<0.05).随时间变化,实验组左侧caspase-12和Bak的含量逐渐升高,实验组右侧逐渐降低.结论 咀嚼力降低会影响髁突软骨中caspase-12和Bak的含量,引起内质网应激介导的凋亡反应,内质网途径是髁突软骨细胞凋亡的重要途径之一.

    • 摘要: 目的 探讨Bcl-2和Bak基因沉默对MG-63细胞凋亡、成骨活动以及钙离子浓度的影响.方法 构建沉默Bcl-2和BaK表达的腺病毒载体,scramble RNA载体转染MG-63细胞,采用MTT检测转染后细胞存活率的情况,并应用ALP观察转染后细胞成骨活动情况以及流式细胞术检测钙离子的浓度间接测定细胞凋亡情况.结果 Bcl-2基因沉默后48 h相对对照组,细胞存活率降低,成骨活动减少,钙离子浓度升高;BaK基因沉默后细胞增存活率升高,成骨活动增加,细胞内钙离子浓度降低,而Bcl-2+BaK联合沉默组与对照组在各观察指标上差异无显著性.结论 Bcl-2和BaK基因沉默对人成骨肉瘤MG-63细胞凋亡,钙离子浓度变化以及成骨活动产生影响,表明其可能与骨质疏松的发病有密切的关联.%Objective To explore the effects of Bcl-2 and BaK gene silencing on cell apoptosis ,osteogen-esis activity and free Ca2+concentration of MG-63 cell lines. Methods The siRNA sequences targeted Bcl-2 and BaK respectively were designed;Bcl-2 and BaK silencing adenovirus vector scramble RNA vector and empty vec-tor were constructed to transfect MG-63 cell lines. MTT method was used to examine cell viability;ALP and flow cytometry were conducted to observe osteogenesis activity and free Ca2+concentration. Results Bcl-2 gene silenc-ing decreased cell viability,reduced osteogenesis activity and increased free Ca2+ concentration when compared with controls but BaK gene silencing had the opposite effects. The effect of Bcl-2+BaK gene silencing on cell was similar to the empty control. Conclusions Cell apoptosis,osteogenesis activity and free Ca2+concentration of MG-63 change following Bcl-2 and BaK gene silencing,implicating their roles in osteoporosis.

    • 摘要: 利用PCR技术扩增日本吸血虫凋亡相关基因BAK(Bcl-2 homologous antagonist/killer),并构建重组质粒.应用Real-time PCR技术分析在日本血吸虫非易感宿主大鼠和易感宿主小鼠来源、不同发育阶段虫体中BAK基因的表达状况.结果显示,克隆获得日本血吸虫(Schistosoma japonicum,Sj)BAK编码基因SjBAK,其ORF含2142bp,编码713个氨基酸,理论分子量为79.3 kDa,理论等电点为4.9.结构域分析表明该基因编码蛋白具有至少3个BH结构域,属于Bcl家族成员.Real-time PCR结果显示该基因在大、小鼠来源4个不同发育阶段虫体均有表达,其中大鼠来源虫体SjBAK的表达水平都高于小鼠来源虫体,在感染后32 d和42 d表达差异具有显著统计学意义(P<0.05).由此可推测日本血吸虫凋亡相关基因SjBAK在血吸虫生长发育中可能发挥重要的作用.%The objectives of the present study were to clone the apoptosis related gene ofSchistosoma japonicumSjBAK(Bcl-2 homologous antagonist/killer), analyze its expression in schistosome worms and possible role in the development of schistosome. The full-length sequence ofSjBAK was cloned in PCR and the recombinant plasmid containingSjBAK was constructed. The expression of SjBAK inS. japonicum at different development stages from non-susceptible host rats and susceptible host mice were analyzed using Real-time PCR. The ORF of theSjBAK gene contained 2142 bp encoding 713 amino acids with the theoretical molecular weight of 79.3 kDa and theoretical isoelectric point at 4.9. Analysis also revealed that the molecule possessed at least three BH domains and belonged to the member of Bcl family. Real-time PCR analysis showed that the gene expressed in all worms at four development stages and the expression level in worms from rats was higher than those from mice. Signifi cant difference inSjBAK expression was observed in worms from two different hosts at 32 d and 42 d post-infection(P<0.05). In conclusion, the apoptosis-related geneSjBAK ofS japonicum may play an important role in the development of Schistosome.

    • 摘要: 目的 探讨雷公藤红素逆转乳腺癌细胞对多柔比星的耐药性及作用机制.方法 将多柔比星耐药MDA-MB-231细胞(MDA-MB-231/R细胞)分为对照组、雷公藤红素组、多柔比星组、雷公藤红素+多柔比星组和雷公藤红素+多柔比星+Bim siRNA组,MTT法检测MDA-MB-231/R细胞活力,流式细胞术检测各组MDA-MB-231/R细胞的凋亡,免疫共沉淀法检测MDA-MB-231/R细胞Bim蛋白与Bak及Bax蛋白的相互作用,Western blot检测MDA-MB-231/R细胞色素C释放和Caspase-3活化.结果 MDA-MB-231/R细胞对多柔比星的半数有效浓度(IC50)显著高于常规MDA-MB-231细胞[(18.4±1.1)g/mL比(1.5±0.1)g/mL,P<0.05];雷公藤红素处理后,多柔比星对MDA-MB-231/R细胞活力抑制率和凋亡诱导率均显著提高,与对照组和多柔比星组比较,差异有统计学意义[(57.3±4.1)%比0、(13.0±1.0)%,(36.5±2.3)%比(1.6±0.2)%、(8.8±0.6)%,P均<0.051;雷公藤红素处理能显著上调MDA-MB-231/R细胞Bim蛋白表达水平[(0.51±0.04)比(0.11±0.01)、(0.13±0.01),P均<0.05];用Bim siRNA阻断Bim蛋白上调后,雷公藤红素对多柔比星的协同抗肿瘤效应受到明显抑制[(20.5±2.1)%比(57.3±4.1)%,(12.4±1.1)%比(36.5±2.3)%,P<0.05];雷公藤红素处理后,与MDA-MB-231/R细胞Bim蛋白结合的Bak、Bax蛋白表达水平显著提高[(0.29±0.02)比(0.04±0.01)、(0.05-±0.01),(0.35±0.02)比(0.05±0.01)、(0.04±0.01),P均<0.05)];雷公藤红素处理后,多柔比星对MDA-MB-231/R细胞色素C的释放率和Caspase-3的活化率显著提高[(0.57±0.05)比(0.11±0.02)、(0.06±0.01),P均<0.05;(0.43±0.04)比(0.09±0.02)、(0.03±0.01),P均<0.05].结论 雷公藤红素可通过上调Bim蛋白表达,提高多柔比星耐药乳腺癌细胞对多柔比星的敏感性.

    • 摘要: 目的:探讨核因子-κB( Nuclear factor kappa B,NF-κB)介导的促凋亡蛋白Bak( Bcl-2 associated K protein gene, Bak)及TNF-α的表达在溃疡性结肠炎( Ulcerative colitis,UC)发病中的作用机制。方法:选购80只清洁级SD大鼠,分为模型组和对照组。 UC大鼠模型制造采用“三硝基苯磺酸+乙醇”方法,模型制造成功后观察、评估结肠黏膜的大体形态及组织学变化。用免疫组织化学及RT-PCR法检测模型组与对照组中的NF-κB、Bak、TNF-α的表达水平,并分析相互之间关系。结果:UC大鼠模型制造成功率为97%。模型组固有层和黏膜层内炎性细胞浸润较对照组明显增多( P<0.01);NF-κB、TNF-α蛋白及mRNA表达水平模型组较对照组明显升高(P<0.01);Bak蛋白在模型组的炎细胞中表达明显低于对照组(P<0.01),而在结肠黏膜上皮细胞中的水平与对照组无明显差异( P>0.05)。 NF-κB、TNF-α蛋白阳性细胞百分数及mRNA水平随模型组的组织学等级升高而增强,而Bak蛋白阳性细胞百分数是减弱的( P<0.05);且模型组中NF-κB与Bak蛋白的表达水平呈负相关( r=-0.793,P<0.01),NF-κB与TNF-α蛋白表达成正相关(r=0.892,P<0.01)。结论:NF-κB介导的促凋亡蛋白Bak的表达参与了UC的发生、发展。%Objective:To study the expression levels of nuclear factor kappa B, Bcl-2 associated K and TNF-αproteins to discuss the effects of NF-κB and Bak proteins in the pathogenesis of UC.Methods:Eighty clean grade of adult Sprague-Dawley( SD) rats were used,male and female in half and then rando mly selected sixty as the model group,another twenty as the control group.SD rats model were manufactured by a compound method:Trinitrobenzene sulfonic acid ( TNBS )+ethanol.We observed and assessed colonic mucosa by the general morphology and histological changes.To applicated immunohistochemistry and RT-PCR methods to detected the protein and mRNA expression levels of NF-κB,Bak and TNF-αin the model groups and the control group and to analysed their relationships.Results:The successful rate of making model was 97%.The number of inflammatory cells in the model groups more than the control(P0.05).The expression levels of NF-κB,TNF-αincreased as the histological grade increased(P<0.05),however,the expression level of Bak decreased(P<0.05).NF-κB in colonic mucosa of rats with UC had a significantly positive correlation with that of TNF-α(r=0.892,P<0.01),and negatively correlated with that of Bak(r=-0.793,P<0.01).Conclusion:The levels of NF-κB and Bak may be related to the occurrence and development of UC.

    • 摘要: 目的 探究N-乙酰半胱氨酸萝卜硫素(D,L-sulforaphane N-Acetyl-L-cysteine,SFN-NAC)诱导前列腺癌细胞凋亡的分子机制. 方法 将前列腺癌细胞DU145和PC-3分别培养在6孔板中,当细胞密度达到70%-80%时,分别随机分为对照组(不加SFN-NAC)和加药组(加入终浓度为30 μmol/L的SFN-NAC),继续在37°C培养24 h.利用Annexin Ⅴ异硫氰酸荧光素/碘化丙啶(FITC/PI)双染流式细胞术检测细胞凋亡程度;利用Western blot检测Bak、Bax、Bcl-2和cleaved Caspase-3蛋白表达水平. 结果 与对照组相比,加药组的细胞凋亡率增高(P<0.01).加药组细胞Bak蛋白表达较对照组升高(P<0.05);Bax蛋白表达也有少量增加,但与对照组相比,无统计学差异(P >0.05);Bcl-2蛋白表达下降(P<0.05);Bax/Bcl-2比值增加(P<0.01);细胞cleaved Caspase-3表达水平升高(P<0.01). 结论 N-乙酰半胱氨酸萝卜硫素通过上调Bak、下调Bet-2蛋白表达,活化Caspase-3,诱导前列腺癌细胞凋亡,说明N-乙酰半胱氨酸萝卜硫素通过线粒体途径发挥抗癌作用.

    • 摘要: 目的:探讨欧前胡素逆转宫颈癌细胞对顺铂抵抗性的机制。方法将顺铂耐药Hela细胞(Hela/R细胞)按对照组、欧前胡素组、顺铂组、欧前胡素+顺铂组及欧前胡素+顺铂+Bim siRNA组进行分组后,采用MTT法检测Hela/R的细胞活力,流式细胞术检测Hela/R细胞的凋亡,免疫共沉淀实验检测Hela/R细胞Bim蛋白与Bak及Bax蛋白的相互作用,Western blot实验检测Hela/R细胞色素c的释放和caspase-3的活化。结果 Hela/R细胞对顺铂的IC50显著高于常规Hela细胞(P<0.05)。欧前胡素处理可诱导Hela/R细胞Bim蛋白的上调及其与Bak、Bax蛋白的相互作用。欧前胡素+顺铂组对Hela/R细胞活力的抑制率和凋亡诱导率显著高于顺铂组和欧前胡素+顺铂+Bim siRNA组。欧前胡素+顺铂组对Hela/R 细胞色素c 的释放和caspase-3的活化显著高于顺铂组和欧前胡素+顺铂+Bim siRNA组。结论欧前胡素通过上调Bim蛋白的表达提高顺铂耐药宫颈癌细胞对顺铂的敏感性。

    • 摘要: Objective:To investigate the application of improved technology TLIF combined passing pedicle screw fixation,interbody fusion( BAK) to treat prolapse of lumbar disc herniation and lumbar instability. Methods:Application of improved technology TLIF combined with posterior pedicle screw and interbody fusion and fusion treat 65 cases of low back paln and radicular paln with unilateral lower limb prolapse of lumbar disc herniation and lumbar instability patients, the patients were examined visual analogue paln score ( VAS ) before and after operation, to survey department of intervertebral angle,the whole lumbar lordosis angle,the edge of the disc space height measurement and surgical sat-isfaction,to evaluate the therapeutic effect. Results:To follow-up 12 ~36mo, 22mo average. VAS before operation and the department of intervertebral angle, whole lumbar lordosis angle and Intervertebral height of the tralling edge,respectively,7. 8±1. 8 points and 3. 4±0. 3°,40±0. 6°,8±0. 5 mm,the first check:2.9±1.2 points(P=0.0082,P<0.01)and 12.6±0.4°(P=0.0075,P<0.01)、52°±0. 5°(P=0. 0061,P<0. 01),12±0. 3mm(P=0. 0054,P<0. 01). Among 65 patients,60 cases surgery satisfaction survey more than 40 points ( 92 . 3%) , and they considered excellent surgical results. Complication of improper operation and Equipment problems didn ’ t happen. Conclusion:posterior improved technology TLIF combines with the pedicle screw fixation、 interbody fusion and fusion surgery is a safe and effective surgical methods to treat Prolapse Dype Lumbar Disc Herniation and lumbar instability.%目的::探讨应用改良TLIF技术结合后路经椎弓根钉棒内固定、椎间融合器( BAK)植骨融合术治疗脱出型腰椎间盘突出症并腰椎不稳症。方法:应用改良TLIF技术结合后路椎弓根钉棒椎间融合器植骨融合术治疗65例腰痛并单侧下肢根性疼痛的脱出型腰椎间盘突出症并腰椎不稳症病人,术前和术后对病人进行视觉疼痛评分( visual analogue pain score,VAS),椎间系角度、整个腰椎前凸角度、椎间隙后缘高度测量和手术满意度问卷调查,评价治疗效果。结果:随访12~36mo,平均22mo。术前VAS和椎间系角度整个腰椎前凸角度椎间隙后缘高度,分别是7.8±1.8分和3.4±0.3°、40±0.6°、8±0.5mm,随访时为2.9±1.2分(P=0.0082,P<0.01)和12.6±0.4°(P=0.0075,P<0.01)、52°±0.5°(P=0.0061,P<0.01)、12±0.3mm(P=0.0054,P<0.01)。65例病人中60例手术满意度问卷调查高于40分(92.3%)认为手术效果优良。没有发生因操作不当及器械问题引起的并发症。结论:后路改良TLIF技术结合经椎弓根钉棒内固定、椎间融合器植骨融合术是一种安全有效的治疗脱出型腰椎间盘突出症并腰椎不稳症的手术方法。
