    • 摘要: 研究以球等鞭金藻(Isochrysis galbana)为研究对象, 分析在不同浓度氟苯尼考暴露下, 球等鞭金藻的生长、叶绿素、类胡萝卜素和脂肪酸含量的变化.结果表明, 氟苯尼考对球等鞭金藻的生长呈"低促高抑"作用,其72h-EC50为17.12 mg/L;随着暴露浓度(≥1 mg/L)增加,细胞内叶绿素含量显著下降,光合作用受到抑制,而类胡萝卜素含量显著上升.脂肪酸和类胡萝卜素共同参与藻细胞的防御机制, 氟苯尼考(< 1 mg/L)暴露引起细胞内总脂肪酸含量显著降低.研究探究了水产养殖环境中抗生素残留的生态风险, 为水产养殖过程中抗生素的科学合理使用提供了理论依据.

    • 摘要: Effect of IAA at 0.1~5 and 0.1~3 mg/L on the maximal photochemical efficiency of PSⅡ (Fv/Fm),relative electron transport rate (rETR),photochemical quenching (qP),cell density,chlorophyll content,total lipid content and fatty acid composition of Isochrysis galbana and Ochromonas neopolitana was investigated,respectively.The results showed that the optimal IAA concentration for the photosynthesis and growth of I.galbana and O.neopolitana was 0.1 and 0.25 mg/L,respectively,at which,Fv/Fm,rETR,qP,cell density and chlorophyll content of two microalgae were all higher than those of control and those at other concentrations.Compared with control,the cell density and chlorophyll content of I.galbana increased by 32.6% and 37.1%,respectively.Its total lipid content was higher than that at other concentrations but similar to that of control.For O.neopolitana,the cell density and chlorophyll content at 0.25 mg/L increased by 48.7% and 34.4%,respectively,however the high-est total lipid content of this alga was detected at 0.5 mg/L,which was similar to that at 0.25 mg/L.For fatty acid composition,the 0.1 mg/L IAA enhanced the synthesis of DHA,but not other fatty acid components (14:0,16:0,16:1n-7,18:1n-9).The 0.25 mg/L IAA enhanced the synthesis of both DHA and other polyunsaturated fatty acids of O.neopolitana.Our findings indicated that the optimal IAA concentration for the growth and oil accumulation of I.galbana and O.neopolitana was 0.1 mg/L and 0.25 mg/L,respectively,which provided the theoretic basis for large scale cultivation of two microalgae.%本文研究了0.1~5 mg/L的-3吲哚乙酸(IAA)对球等鞭金藻(Isochrysis galbana)和0.1~3 mg/L的IAA对棕鞭藻(Ochromonas neopolitana)的叶绿素荧光参数(PSⅡ最大光能转化效率Fv/Fm、相对电子传递速率rETR、光化学淬灭qP)、细胞密度、叶绿素含量、总脂含量及脂肪酸组成的影响.研究表明,球等鞭金藻和棕鞭藻进行光合作用和生长的最适IAA浓度分别为0.1和0.25 mg/L.此条件下,2株藻的Fv/Fm、rETR、qP、细胞密度和叶绿素含量均显著高于对照组和其它处理组.其中,球等鞭金藻的细胞密度和叶绿素含量分别比对照组增加了32.6%和37.1%,总脂含量显著高于其它处理组,但与对照组差异不显著(P>0.05);棕鞭藻的细胞密度和叶绿素含量在IAA浓度为0.25 mg/L时分别比对照组增加了48.7%和34.4%,总脂含量在IAA浓度为0.5mg/L时显著高于对照组和其它处理组,但与0.25 mg/L处理组差异不显著(P>0.05).0.1 mg/L的IAA浓度对球等鞭金藻DHA(二十二碳六烯酸)的合成有促进作用,但对其它各脂肪酸(14:0、16:0、16:1n-7和18:1n-9)含量均无显著影响;0.25 mg/L的IAA浓度促进了棕鞭藻DHA和多不饱和脂肪酸的合成.研究结果表明,球等鞭金藻和棕鞭藻进行生长和油脂积累的最适IAA浓度分别为0.1和0.25mg/L,此结果为2株藻的大规模培养提供了理论依据.

    • 摘要: Isochrysis galbana is an excellent unicellular bait for the larvae of many aquaculture animals. Thus, screening for I. galbana strains with rapid growth rate and high content of docosahexaenoic acid (DHA) and ei-cosapentaenoic acid (EPA) is very important for the breeding of aquaculture animals. However, most of the I. galbana strains in China have been derived from a common ancestor called I. galbana OA-3011, and different strains of I. galbana have been developed after a long duration of geographical isolation. Whether the traits of these strains have changed or not remains unknown. In order to evaluate the status of the phenotypic traits of dif-ferent geographic isolates for future selective breeding, comparative analysis of five strains of I. galbana (FACHB-861, OA3011-QD, OA3011-LK, OA3011-AY, and OA3011-LZ) with regard to their taxonomy, growth, and fatty acid composition was conducted. Different strains were cultured under identical conditions for six days until they reached a stationary phase. Genomic DNA was extracted for polymerase chain reaction sequencing of the 18S rDNA gene. The OD660 was recorded daily for plotting a growth curve. When the cultures reached statio-nary phase, they were harvested by high-speed centrifugation for biomass determination and fatty acid analysis by using gas chromatography. Phylogenetic analysis of the 18S rDNA gene suggested that FACHB-861 was distantly related with the OA-3011 strains. OA3011-LK, OA3011-AY, and OA3011-LZ had identical 18S rDNA sequences, but the 18S rDNA sequence of OA3011-QD differed from that of the other OA-3011 strains at one position, re-sulting in a single base substitution. The growth curve showed that the five strains reached stationary phase at dif-ferent times. FACHB-861 showed a distinct pattern of growth, unlike the other four OA-3011 strains. The final biomass concentrations were significantly different between I. galbana FACHB-861 and OA-3011 strains as well as among OA3011-LK, OA3011-AY, and OA3011-LZ (P<0.05), whereas the growth rates were not significantly different among the strains (P>0.05). The total fatty acid content of FACHB-861 and OA3011-LZ were signifi-cantly lower than that of OA3011-LK, OA3011-QD, and OA3011-AY (P<0.05). The fatty acid profile of FACHB-861 was remarkably different from that of the OA-3011 strains. However, a conspicuous variation in fatty acid compositions was also observed among OA-3011 strains, indicating a wide and frequent quantitative pheno-typic variation among the strains of I. galbana. These variations resulting from geographic isolation might provide a feasible method for the selective breeding of I. galbana strains. According to the screening criteria such as growth rate or DHA, EPA, and polyunsaturated fatty acid content, our preliminary analysis showed that OA3011-LK, which has the highest growth rate, and OA3011-QD, which contains the highest amount of DHA, are promising candidates for further screening.%球等鞭金藻作为一种优良的单细胞生物饵料具有选育价值.目前我国水产养殖上使用的大多数球等鞭金藻品系都来源于同一株藻株Isochrysis galbana OA-3011,经过多年的地理隔离形成了不同的品系,为了揭示地理隔离对球等鞭金藻表型性状的影响,从而为球等鞭金藻的选育工作提供依据,我们对5个品系的球等鞭金藻(I.gal-bana FACHB-861,OA3011-QD,OA3011-LK,OA3011-AY和OA3011-LZ)的种属关系、生长和脂肪酸组成进行了比较分析.实验在相同条件下对5个品系球等鞭金藻培养6 d,提取基因组DNA并PCR扩增测定18S rDNA基因序列,每天定时记录各品系的OD660,绘制生长曲线,在进入稳定期后收获并进行生物量的测定及脂肪酸的气相色谱分析.18S rDNA基因序列的进化分析表明,OA3011-LK、OA3011-AY和OA3011-LZ具有一致的序列,而OA3011-QD因1个碱基的差异与其他OA3011出现了分离,FACHB-861与OA3011品系亲缘关系较远;对生长和脂肪酸进行对比分析后,结果显示,5个品系的球等鞭金藻在不同的时间达到稳定期,其中FACHB-861表现出了与其他品系明显不同的生长模式.FACHB-861和OA-3011品系的生物量具有显著差异,OA3011-LK、OA3011-AY和OA3011-LZ之间也有显著差异(P<0.05),但特异生长率各品系间无显著差异(P<0.05).FACHB-861和OA3011-LZ的总脂肪酸(TFA)含量显著低于OA3011-LK、OA3011-QD和OA3011-AY(P<0.05).FACHB-861的脂肪酸各成分与OA-3011各品系明显不同.然而,在OA-3011各品系间脂肪酸组成也有明显差异,这表明球等鞭金藻存在广泛和频繁的数量性状表型差异.总之,地理隔离造成的这些差异为球等鞭金藻的选育提供了可行性.根据生长速度或DHA、EPA、多不饱和脂肪酸含量等筛选标准初步分析显示,可对OA3011-LK,OA3011-QD进行进一步的选育.

    • 摘要: 本文研究主要探索植物中的丙氨酸对球等鞭金藻生长的影响实验,为了对比实验研究成果,本文拟对卤地菊中两种氨基酸对球等鞭金藻生成的影响进行对比分析,从而确定陆地植物浸提液对球等鞭金藻生长的权重影响.研究结果表明,卤地菊水浸提液对球等鞭金藻的生长具有显著的促进作用;卤地菊中丙氨酸对球等鞭金藻具有显著的促进作用.通过进一步计算可知,在本文实验条件下,丙氨酸对球等鞭金藻促进的最佳浓度为0.20g/L,其在卤地菊水浸提液中的最大权重系数为15.17%;甘氨酸低浓度时具有促进作用,高浓度呈现出抑制作用,较高浓度时在1~5d有促进作用,从第5天开始甘氨酸对球等鞭金藻的生长具有不同程度的抑制作用,呈现"低促高抑"的特点.本研究结果可以为控制水域污染和海产品养殖提供实验基础.

    • 摘要: The growth, cellular protein content and fatty acid profile in micro-algae Isochrysis galbana were valuated when the culture was supplemented with 0, 0.5, 1.0, 1.5 and 2.0 mg/mL NaHCO3 respectively. Then the protein content and fatty acid profile in cell at different growth phase were also investigated when the culture medium was added with 1.5 mg/mL NaHCO3. The results indicated there were significant effects of NaHCO3 supplementation on growth, cellular protein content and fatty acid profiles. The treatment supplemented with 1.5 mg/mL NaHCO3 displayed the highest cell density and special growth rate. The cellular protein content decreased significantly along with the increasing supplementation of NaHCO3 in culture medium. The monounsaturated fatty acids increased significantly while the polyunsaturated fatty acids decreased significantly along with the increase of NaHCO3 supplementation. There were significant differences in cellular protein content and fatty acid profile among the cells which were at a different growth phase when the cells were cultured in the medium supplemented with 1.5 mg/mL NaHCO3. The saturated fatty acids and monounsaturated fatty acids increased significantly while the polyunsaturated fatty acids decreased significantly along with the culture duration. Therefore it was suggested that NaHCO3 supplementation in culture medium was helpful for improving the micro-algae quality for bio-diesel production.%本文研究了培养液中添加0、0.5、1.0、1.5和2.0 mg/mL NaHCO3对球等鞭金藻(Isochrysis galbana)生长、细胞蛋白质含量及脂肪酸组成的影响,并进一步探讨了添加适宜NaHCO3后不同培养阶段对细胞营养组成的影响。结果表明:NaHCO3对球等鞭金藻的生长和细胞营养价值有显著影响。批次培养模式下培养液中添加1.5 mg/mL NaHCO3可以获得最大细胞密度和比生长率。球等边金藻细胞中蛋白质含量随培养液中NaHCO3添加量的增大而显著减少;而细胞中得单不饱和脂肪酸(MUFA)显著增加,多不饱和脂肪酸(PUFA)显著降低。在添加1.5 mg/mL NaHCO3的培养条件下,不同培养阶段的藻细胞蛋白和脂肪酸组成也有显著变化,随着培养时间的延长,SFA和MUFA含量逐渐增加,PUFA含量则显著减少。培养液中添加NaHCO3有助于提升球等鞭金藻作为生物柴油原料的品质。

    • 摘要: 本文研究了批次培养模式下培养基中分别添加0.018 mol/L不同碳源(NaHCO3,C6H12O6,CH3COONa和CH3CH2COONa)对球等鞭金藻(Isochrysis galbana)生长、色素含量、蛋白质含量和脂肪酸组成的影响.结果表明:在盐度26,湿度25℃,光照3000 lux条件下,f/2培养液中添加不同碳源对球等鞭金藻的生长和细胞组成有显著影响.不同碳源对球等鞭金藻促生长效果依次为:葡萄糖>碳酸氢钠>空白对照>丙酸钠>乙酸钠.培养液中添加各种碳源均不同程度上降低了球等鞭金藻细胞蛋白质含量.添加碳酸氢钠、乙酸钠和丙酸钠组藻叶绿素a和类胡萝卜素含量均显著降低,而添加葡萄糖组藻细胞中两种色素含量显著增加.添加碳源改变了球等鞭金藻细胞脂肪酸组成,单不饱和脂肪酸(MUFA)含量均显著增加,而多不饱和脂肪酸(PUFA)均显著降低.上述研究表明葡萄糖是促进球等鞭金藻生长的高效碳源,培养液中添加葡萄糖是球等鞭金藻高密度培养的有效策略.

    • 摘要: 微藻高效培养是微藻生物能源开发利用的关键和前提,而在营养充足的培养条件下生长迅速但较易受到环境污染和影响,因此微藻生长过程中对其生长状况进行监测意义重大.高光谱成像技术同时拥有丰富物质品质信号的优点和图像包含丰富品质分布空间信息的优点,可为微藻的快速无损检测提供新的方法和手段.分别采集小球藻、球等鞭金藻和螺旋藻三种微藻各45个样本的高光谱图像,并提取样本感兴趣区域(ROI)的平均光谱.利用连续投影算法(SPA)波长优选之后,取30个建模集样本的光谱数据与其相应的生物量建立多元线性回归(MLR)模型,对15个预测集样本的生物量进行预测,小球藻、球等鞭金藻和螺旋藻预测相关系数(r)分别为0.950,0.969和0.961,预测均方根误差(RMSEP)为0.010 2,0.010 7和0.017 1,获得了较好的预测精度.最后,用所建MLR模型对预测集图像上每个像素点的生物量加以预测,采用Matlab图像编程处理将不同的生物量用不同的颜色表示,最终以伪彩图的形式实现藻液生物量的可视化.研究结果表明,高光谱成像技术对小球藻和螺旋藻藻液生物量的可视化效果较好,对球等鞭金藻的预测效果还需要进一步改进.本研究为实现微藻生长信息的快速获取和进一步开展微藻生物质能源利用奠定了一定的研究基础.

    • 摘要: 本研究以变态存活率、产卵量、成体头胸部长和体长为指标,研究了球等鞭金藻(Isochrysis galbana)和海洋原甲藻(Prorocentrum micans)不同浓度和配比对太平洋纺锤水蚤(Acartia pacifica)生长发育和产卵量的影响,以期探索培养太平洋纺锤水蚤最适合的饵料配方.实验结果表明:在饵料浓度(碳含量,下同)为1.70μg/cm3,配比为2∶1时,其变态存活率最高,可达到92.62%;在饵料浓度为4.25μg/cm3,配比为1∶2时,产卵量最高,雌体平均日产卵量可达47.1±2.7个;在饵料浓度为1.70μg/cm3,配比为2∶1时,成体头胸部长和体长均最长,分别为932.9、1 252.3μm.太平洋纺锤水蚤生长和发育的最优饵料组合为球等鞭金藻与海洋原甲藻浓度为1.70μg/cm3,配比为2∶1.
