    • 摘要: 目的 探讨乳腺癌临床病理特征和临床预后与血清Leptin、蛋白激酶Cε(PKCε)的相关性.方法 选取河北大学附属医院2017年11月至2018年2月乳腺癌患者40例作为观察组,50例体检健康女性作为对照组;对患者进行随访2年,详细记录患者预后质量;采集受试者空腹静脉血检测血清Leptin、PKCε水平;采用Spearman分析Leptin、PKCε水平与患者临床病理特征的相关性;绘制卡米尔生存曲线分析不同Lep-tin、PKCε水平情况患者预后质量;采用Logistics回归模型分析血清Leptin、PKCε水平对乳腺癌患者预后的影响.结果 观察组血清Leptin、PKCε水平均明显高于对照组(P<0.05);不同TNM分期、病灶区最大径和腋窝淋巴结转移患者血清Leptin、PKCε水平比较差异有统计学意义(P<0.05),随着TNM分期升高、病灶区最大径增大及腋窝淋巴结转移出现,患者血清Leptin、PKCε水平呈明显升高状态(P<0.05);患者血清Leptin、PKCε水平与TNM分期、病灶区直径、腋窝淋巴结转移呈正相关(r=0.675~0.743,P<0.05);死亡组患者血清Leptin、PKCε水平均高于存活组(P<0.05),高水平Leptin和高水平PKCε患者生存率明显低于低水平Leptin和低水平PKCε患者(P<0.05);血清高水平Leptin和高水平PKCε是影响乳腺癌患者预后质量的独立危险因素(P<0.05).结论 乳腺癌患者血清Leptin、PKCε水平与TNM分期、病灶区直径、腋窝淋巴结转移呈正相关,且高水平Leptin和高水平PKCε是影响患者预后质量的独立危险因素.

    • 摘要: 目的 探讨Leptin对体外培养的缺氧状态下视网膜祖细胞(RPCs)增殖、分化的影响及细胞中PTEN蛋白表达的变化。方法 原代培养大鼠RPCs,不同浓度Leptin作用RPCs细胞,分为:常氧培养组(Control组)、缺氧培养(Hypoxia)+0、0.3、1.0、3.0、10、30 nmol/L Leptin组,分别培养至12、24、48 h,CCK-8法检测细胞增殖活性。将原代培养的RPCs细胞分为:Control组、Hypoxia组、Hypoxia+Leptin组,培养48 h后,转换为分化培养基继续培养6 d,采用免疫荧光染色法对GFAP阳性细胞比例和β-tubulin Ⅲ阳性细胞比例进行统计分析。Control组、Hypoxia组、Hypoxia+Leptin组细胞培养48 h后,采用Western blot法检测各组RPCs中PTEN蛋白表达水平。结果 原代培养的P0代RPCs为圆形悬浮生长,培养2 d后细胞可聚集成神经球。低浓度Leptin(0.3、1.0、3.0 nmol/L)作用RPCs 48 h,细胞增殖活性较Control组及Hypoxia组增强,RPCs细胞增殖活性随Leptin浓度增高呈递增趋势,其中,3.0 nmol/L Leptin作用48 h细胞增殖活性最强(P0.05),而PTEN蛋白表达量较对照组显著下调(P<0.05)。结论 低浓度Leptin可促进缺氧状态下大鼠RPCs细胞增殖,抑制细胞中PTEN蛋白表达,对细胞分化无明显影响。

    • 摘要: BACKGROUND Unexplained infertility(UI)is usually used for any heterosexual couple who failed to have a successful clinical pregnancy without identifying clear causes after they undergo all standard fertility tests.Evidence shows that leptin is one of the most accurate biomarkers for UI.Nevertheless,conflicting results regarding leptin levels in women with UI have been reported.AIM To find the serum leptin levels in women with UI.METHODS All studies written in English and conducted before April 30,2021 from PubMed/MEDLINE,Embase,Cochrane Central Register of Controlled Trials,ClinicalTrials.gov,Google Scholar,OpenGrey,OATD,and the infertility conference abstract were included.Studies were found eligible if they provided the mean and standard deviation of leptin for the case group and control group.The quality assessment of individual studies was evaluated using the Joanna Briggs Institute Quality Assessment Tool.Data synthesis and statistical analysis were done using STATA software version 16.RESULTS A total of 378 studies were reviewed,and just six studies that fulfilled the eligibility criteria were included in this meta-analysis.The pooled result showed that leptin levels were significantly higher in women with UI compared to fertile women,with a standardized mean difference of 0.97(95%confidence interval:-0.49-2.43).However,heterogeneity across studies was highly significant(P<0.00001;I2=98.8%).CONCLUSION The results of this study suggest that leptin levels are elevated in women with UI compared with fertile women;hence,leptin could be a potential biomarker for UI in women,and it may be useful for identifying women with a high risk of infertility.

    • 摘要: BACKGROUND Insulin resistance(IR)and impaired energy expenditure(IEE)are irreparable metabolic comorbidities in schizophrenia.Although mechanism(s)underlying IR and IEE remains unclear,leptin and fatty acid signaling,which has profound influence on insulin secretion/sensitivity,glucose metabolism and energy expenditure,could be disrupted.However,no association of plasma leptin with erythrocyte membrane fatty acids,body mass index(BMI),and psychotic symptoms in the same cohort of untreated patients with first-episode psychosis(FEP)or medicated patients with chronic schizophrenia(CSZ)is presented before.These studies are crucial for deciphering the role of leptin and fatty acids in the development of IR and IEE in schizophrenia.AIM To determine the association between plasma leptin,erythrocyte membrane fatty acids,particularly,saturated fatty acids(SFAs),BMI and psychotic symptoms in patients with FEP and CSZ.METHODS In this study,twenty-two drug naive patients with FEP,twenty-one CSZ patients treated with atypical antipsychotic drugs,and fourteen healthy control(CNT)subjects were analyzed.Plasma leptin was measured using sandwich mode enzyme-linked immunosorbent assay.Erythrocyte membrane SFAs were measured using ultrathin capillary gas chromatography.BMI was calculated by using the formula:weight(kg)/height(m^(2)).Psychiatric symptoms were evaluated at baseline using brief psychiatric rating scale(BPRS),and positive and negative syndrome scale(PANSS).The total BPRS scores,positive and negative symptom scores(PANSS-PSS and PANSS-NSS,respectively)were recorded.Pearson correlation coefficient(r)analyses were performed to find the nature and strength of association between plasma leptin,PANSS scores,BMI and SFAs,particularly,palmitic acid(PA).RESULTS In patients with FEP,plasma leptin not BMI was significantly lower(P=0.034),whereas,erythrocyte membrane SFAs were significantly higher(P<0.005)compared to the CNT subjects.Further,plasma leptin showed negative correlation with erythrocyte membrane SFAs-PA(r=-0.4972,P=0.001),PANSS-PSS(r=-0.4034,P=0.028),and PANSS-NSS(r=-0.3487,P=0.048).However,erythrocyte membrane SFAs-PA showed positive correlation with PANSS-PSS(r=0.5844,P=0.0034)and PANSS-NSS(r=0.5380,P=0.008).In CSZ patients,plasma leptin,BMI,and erythrocyte membrane SFAs,all were significantly higher(P<0.05)compared to the CNT subjects.Plasma leptin showed positive correlation with BMI(r=0.312,P=0.032)but not with PANSS scores or erythrocyte membrane SFAs-PA.However,erythrocyte membrane SFAs-PA showed positive correlation with PANSS-NSS only(r=0.4729,P=0.031).Similar changes in the plasma leptin and erythrocyte membrane SFAs have also been reported in individuals at ultrahigh risk of developing psychosis;therefore,the above findings suggest that leptin-fatty acid biosynthesis could be disrupted before the onset of psychosis in schizophrenia.CONCLUSION Disrupted leptin-fatty acid biosynthesis/signaling could be an early manifestation of metabolic comorbidities in schizophrenia.Large-scale studies are warranted to validate the above findings.

    • 摘要: BACKGROUND Kawasaki disease(KD)is an acute self-limited vasculitis with a predilection for coronary arteries.Children with KD may have altered lipid metabolism and abnormal lipid profiles that may last for prolonged periods.However,there is a paucity of literature on the role of adipocytokines in KD.AIM To estimate the levels of adipocytokines(adiponectin,leptin and resistin)during the convalescent phase of KD.METHODS Twenty children,who had KD at least three years earlier,were enrolled in this study.In addition,20 healthy controls were also enrolled.Clinical and laboratory profiles of patients were obtained from hospital records.Serum adiponectin,leptin and resistin levels were estimated by enzyme-linked immunosorbent assay.RESULTS Mean age of the patients in the study group was 10.15±3 years and the male:female ratio was 1.5:1.Median serum resistin levels in patients with KD(27.77 ng/mL;[IQR:18.66,48.90])were decreased compared to controls(21.20 ng/mL;[IQR:14.80,27.00])(P=0.04).Median serum leptin levels in cases and controls were 1.83 ng/mL;(IQR:1.13,3.80),and 1.10 ng/mL;(IQR:0.41,2.88),respectively(P=0.09).Median serum adiponectin levels were similar in both cases(12.20μg/mL;[IQR:9.76,17.97])and controls(13.95μg/mL;[IQR:11.17,22.58]);(P=0.18).There was no significant difference in all 3 adipocytokines between children with(4/20)and without coronary artery abnormalities(16/20).CONCLUSION Serum resistin levels were significantly elevated in patients with KD during the convalescent phase compared to controls.Serum leptin levels appeared to be higher in patients with KD,although the difference was not statistically significant.Adiponectin levels were similar in both cases and controls.Raised resistin and leptin levels may partially explain lipid perturbations observed during the convalescent phase of KD.

    • 摘要: BACKGROUND The excessive concentration of leptin has negative effects on all aspects of female reproduction.Despite this established relationship,the exact role of leptin in women’s fertility is not clear enough and needs more clarification.AIM To evaluate the serum leptin levels in Sudanese women and to ascertain the relationship between serum leptin levels and unexplained infertility(UI).METHODS A matched(age and body mass index)case-control study was conducted from March 2021 to February 2022.The study samples were 210 women with UI and 190 fertile women of reproductive age who were attending the maternity hospitals and fertility clinics in Khartoum state Sudan.The serum concentration of leptin and other serum biomarkers were determined using enzyme-linked immunosorbent assays.RESULTS The results showed that there was a highly statistically significant difference between the two groups(P0.80),for leptin level,LH/FSH ratio,PRL level,and E2/T ratio,and large(effect size 0.50-≤0.80)for FSH and T.CONCLUSION This study reveals that leptin could be a potential biomarker for UI in Sudanese women and it may be useful for identifying women with a high risk of infertility.

    • 摘要: 目的 观察去卵巢骨质疏松症大鼠骨组织Leptin启动子甲基化,探索补肾健骨中药复方防治绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)的分子机制.方法 切除雌性Wistar大鼠双侧卵巢建立PMOP模型,实验大鼠分为4组:正常组、模型组、补肾健骨中药复方组、仙灵骨葆阳性对照组.给药14周后,X射线骨密度测定仪检测骨密度、显微镜观察股骨头石蜡切片HE染色显微形态结构、质谱法检测Leptin启动子甲基化.结果 ①第1~6腰椎骨密度,模型组与正常组相比显著降低(P<0.01);补肾健骨中药复方组、仙灵骨葆阳性对照组与模型组相比显著升高(P<0.05).②模型组与正常组相比,股骨头骨皮质骨板变薄,骨松质骨小梁明显变细,有些部位破坏、断裂,骨髓腔明显扩大;补肾健骨中药复方组、仙灵骨葆阳性对照组与模型组相比,股骨头显微形态结构病理改变明显改善.③骨组织Leptin启动子-493 bp~-19 bp甲基化水平,模型组与正常组相比CpG19、CpG22.23显著升高(P<0.05);补肾健骨中药复方组(P<0.01)、仙灵骨葆阳性对照组(P<0.05)与模型组相比,CpG19甲基化水平显著降低.结论 补肾健骨中药复方通过降低骨组织Leptin启动子甲基化的分子机制,有效防治PMOP.

    • 摘要: 以猪原代皮下前脂肪细胞为研究材料,检测Leptin介导JAK/STAT信号通路中基因表达水平,旨在阐明Leptin介导JAK/STAT信号通路对脂肪代谢的分子机制.用0和100 ng/mL Leptin分别处理脂肪细胞48 h,油红O染色鉴定脂肪细胞,试剂盒测定细胞中甘油三酯和游离脂肪酸含量,Real-time PCR方法检测皮下前脂肪细胞中JAK/STAT信号通路中基因表达水平.研究结果表明:皮下前脂肪细胞中,Leptin组甘油三酯含量均显著低于对照组(P<0.05),脂肪酸含量均低于对照组;JAK/STAT信号通路中LepR、JAK2、STAT3、CPT-1、ACOX1和PGC-1α基因的表达量均显著高于对照组(P<0.05);LepR、JAK2、STAT3、CPT-1、ACOX1和PGC-1α基因表达量均与甘油三酯和游离脂肪酸含量呈显著负相关(P<0.05).该研究结果表明,Leptin激活皮下前脂肪细胞中JAK/STAT信号通路,上调通路相关基因的表达,促进甘油三酯分解及脂肪酸氧化,降低细胞中甘油三酯和游离脂肪酸含量.

    • 摘要: 试验通过研究不同生长速度阿勒泰羊主要血液生化指标含量和尾脂中瘦素(Leptin)基因的表达水平,探讨阿勒泰羊生长速度与脂肪沉积之间的关系.选取300只出生时间相近的阿勒泰羊公羔,正常饲喂至6月龄,记录日增重,取平均日增重排前10% 和后10% 的公羔各30只,分为生长快速组(HBW)和生长慢速组(LBW),从生长快速组随机选12只分为抑制生长组(饲喂低能量饲粮以抑制其生长,HBW-75%,n=6)和生长快速对照组(饲喂标准饲粮,HBW-100%,n=6),生长慢速组随机选12只分为促进生长组(饲喂高能量饲粮以促进生长,LBW-125%,n=6)和生长慢速对照组(饲喂标准饲粮,LBW-100%,n=6);预饲期7 d,正饲期30 d,试验结束后称重并采集血液,屠宰后采集尾部脂肪.检测血液TG、CK、TSH、T3、T4、S.S、GH、INS、PG、Leptin含量或活性及尾部脂肪Leptin mRNA和蛋白表达量,并观察尾部脂肪细胞形态学变化.结果显示,HBW-100% 和LBW-100% 组初体重与末体重均差异显著(P0.05);LBW-100%和LBW-125% 组初体重差异显著(P0.05).LBW-100% 组的T4、S.S、Leptin、TSH、T3含量显著低于HBW-100% 组(P0.05),Leptin蛋白表达量极显著高于LBW-100% 组(P<0.01).LBW-125% 组Leptin mRNA和蛋白的表达量均显著高于LBW-100% 组(P<0.05).HBW-75% 组Leptin mRNA的表达量极显著低于HBW-100% 组(P<0.01),Leptin蛋白表达量显著低于HBW-100% 组(P<0.05).综上所述,生长快速组与生长慢速组中,Leptin的表达量与体重和体脂有一定关联;在生长慢速组促进生长后体重和体脂显著增加,Leptin的表达量呈上升趋势;在生长快速组抑制生长后体重和体脂减少,Leptin的表达呈下降趋势.改变生长速度对Leptin的表达量会产生影响,从而改变机体的脂肪沉积状况.

    • 摘要: 目的 探究老年难治性精神分裂症患者运用齐拉西酮联合奥氮平的疗效分析.方法 选取118例2017年5月—2019年8月该院收治的老年难治性精神分裂症患者,随机分为观察组(齐拉西酮联合奥氮平治疗)和对照组(单纯奥氮平治疗),每组59例.对比两组治疗效果、Leptin、BDNF、NGF水平改善和不良反应发生情况.结果 观察组治疗总有效率为94.92%,高于对照组77.97%,差异有统计学意义(P0.05),观察组治疗后血清Leptin为(61.24±2.35)ng/mL、BDNF为(39.54±6.74)μg/L、NGF为(45.82±8.65)μg/L,优于对照组,差异有统计学意义(P0.05).结论 老年难治性精神分裂症患者运用齐拉西酮联合奥氮平治疗后,能够提高治疗效果,改善患者机体的相关临床症状和相关实验室指标,且不良反应发生率较低,有利于促进患者疾病恢复.
