    • 摘要: 研究了白色蚊香对蚊的驱避效果及烟尘量.按GB/T 13917.4-2009《农药登记卫生用杀虫剂室内药效试验及评价第4部分:蚊香》和GB/T 13917.10-2009《农药登记卫生用杀虫剂室内药效试验及评价第10部分:模拟现场》试验方法测定五个不同浓度的氯氟醚菊酯白色蚊香对致倦库蚊的室内和模拟现场药效;按GB/T 28015-2011《家用卫生杀虫用品烟尘量试验方法》试验方法对白色蚊香的烟尘量进行试验.结果 表明:浓度为0.08%的氯氟醚菊酯白色蚊香对蚊室内及模拟现场药效为A级,优于其他4个含量,并达到显著水平;该白色蚊香的烟尘量均值为4.7mg/g,达到无烟等级.该白色蚊香具有良好的驱蚊效果,烟尘量低,值得进一步研究应用.

    • 摘要: 目的 了解江津区致倦库蚊幼虫抗药性水平,为蚊虫防治科学用药提供依据。方法 采用幼虫浸渍法,测定野外致倦库蚊幼虫的半数致死浓度(LC50)。结果 致倦库蚊幼虫对高效氯氰菊酯、溴氰菊酯、残杀威、敌敌畏和双硫磷抗性倍数分别为60.24、29.09、4.77、1.50和5.31倍;结论 江津区致倦库蚊幼虫对高效氯氰菊酯为高抗,对溴氰菊酯为中抗,对残杀威和双硫磷为低抗,对敌敌畏为敏感。

    • 摘要: 本研究通过半现场试验探索了背负式喷雾器喷洒Aqualuer?20-20 (有效成份: 菊酯类) 和杀幼剂Altosid?(有效成份: S-烯虫酯) 混合物防制致倦库蚊的效果.结果显示, Aqualuer?20-20, Aqualuer?20-20+Altosid?混合物和对照组喷洒 2 h 后, 成蚊的死亡率分别为 96.29% (92%~100%)、 71.5% ( 57.72%~85.29%) 和4.22% (0.88%~7.57%).单独使用Aqualuer?20-20组、混合使用组和对照组喷洒后24 h死亡率分别为100%、 88.54% (76.23%~100%) 和13.95% (5.12%~22.78%).不同组别成蚊2 h和24 h死亡率差异均有统计学意义 (P<0.05), 与单独使用Aqualuer?20-20组相比, 混合用药提高了幼虫喷洒后2 h的死亡率.本研究为野外蚊虫成虫和幼虫的病媒控制提供了一种合理有效的方法.%The semi-field test was conducted in present study to explore the effect of spraying the mixture of adulticide Aqualuer?20-20 ( active ingredient: permethrin) and larvicide Altosid?( ai: ( S) -methoprene) with backpack ULV sprayer against larvae and adult of Culex quinquefasciatus. The adult mortality 2 h after treatment with Aqualuer?20-20, Aqualuer?20-20+Altosid?mixture, and untreated control groups was 96.29% ( 92%-100%), 71.5% ( 57.72%-85.29%) and 4.22% (0.88%-7.57%), respectively. After 24 h post treatment, the adult mortality was up to 100% in the Aqualuer?20-20 group. In contrast, the mortality in the Aqualuer?20-20+Altosid?mixture and control group were 88.54% (76.23%-100%) and 13.95% (5.12%-22.78%), respectively. There were statistically significant differences in mortality among the three treatments both at 2 h and 24 h post treatment (P<0.05). In larval evaluation, Aqualuer?20-20 alone and Altosid?mixed with Aqualuer?20-20 resulted in comparably significant mortality of larvae 2h after treatment as compared with Aqualuer?20-20 alone. Our study offers an option for combined adult and larval control of mosquitoes in the field.

    • 摘要: [Aim] This study aims to establish and molecularly identify a cell line based on tissues from the newly hatched larvae of the mosquito Culex pipiens quinquefasciatus (Diptera:Culicidae).[Methods] The shredded tissues of the newly hatched larvae of the parathion-resistant strain Shengui of C.pipens quinquefasciatus were cultured in M199 complete medium supplemented with 20% heatinactivated fetal bovine serum,and the primary cells were obtained.The primary cells were treated with trypsin and subcultured into a cell line.The tissue origin of the cell line was identified by observing its biological characteristics under a microscope and comparing DNA amplification fingerprinting and sequencing the ribosome DNA internal transcribed spacer.[Results] The cell line Cxq-1 was well maintained in M199 complete medium and subcultured continually for 12 months.After being frozen in liquid nitrogen,Cxq-1 was successfully resuscitated for several times.DNA amplification fingerprinting polymerase chain reaction clearly distinguished Cxq-1 from two other insect cell lines.Sequencing of ribosome DNA internal transcribed spacer of Cxq-1 indicated that its rDNA-ITS1 and rDNA-ITS2 sequences has higher than 99% identity with those of C.pipens quinquefasciatus registered in the GenBank,confirming the origin of Cxq-1.[Conclusion] The newly established cell line Cxq-1 from newly hatched larvae of C.pipens quinquefasciatus has been confirmed to be a cell line of this mosquito.This cell line provides an important platform for studying the molecular mechanisms of insecticide resistance,mosquito-borne viruses and parasites in the future.%[目的]以致倦库蚊Culex pipiens quinquefasciatus初孵幼虫组织为材料建立细胞系并对其进行分子鉴定.[方法]将实验室筛选的对硫磷抗性的致倦库蚊深桂品系的初孵幼虫剪碎后放入添加20%胎牛血清的改良M199培养基原代培养,并对原代细胞用胰蛋白酶传代培养成细胞系;通过显微镜观察其生物学特性以及DNA扩增指纹图谱和内转录间隔区序列鉴定其组织来源.[结果]该细胞系Cxq-1在M199完全培养基中生长良好,已成功传代培养12个月,并于液氮中冻存,并多次成功复苏.通过DNA扩增指纹图谱鉴定,Cxq-1与实验室培养的其他昆虫细胞DNA指纹图谱的条带各异,表明确为同源昆虫致倦库蚊细胞系.通过内转录间隔区序列测定技术鉴定,Cxq-1的rDNA-ITS1和rDNA-ITS2序列与GenBank中致倦库蚊相应核酸序列的一致性大于99%,表明其来源组织确为致倦库蚊.[结论]新建立的蚊初孵幼虫细胞系Cxq-1确为致倦库蚊细胞系.该细胞系为实验室今后开展杀虫剂抗性分子机制、蚊媒病毒、寄生虫等研究提供了重要平台.

    • 摘要: 采用提取、萃取及柱层析等方法,从山蒟Piper hancei Maxim甲醇提取物的石油醚和氯仿萃取相中分离到6个脂肪链酰胺类化合物,通过核磁共振、质谱并结合相关文献比对,其结构被分别鉴定为已知化合物chingchengenamide A(C1)、N-异丁基-反-2-反-4-癸二烯酰胺(C2)、假荜拨酰胺A(C3)、荜茇宁(C4)、N-p-香豆酰酪胺(C5)和N-反式-阿魏酰酪胺(C6),其中C1为首次从山蒟中获得。利用幼虫浸液法测试了各化合物对白纹伊蚊Aedes albopictus和致倦库蚊Culex pipiens quinquefasciatus幼虫的12 h杀虫活性。结果表明:在20 mg/L下,化合物C1对白纹伊蚊的杀虫活性较高,其校正死亡率为100%,LC50值为5.37 mg/L;化合物C1、C2、C3和C4对致倦库蚊的杀虫活性较高,20 mg/L下的校正死亡率分别为100%、88.5%、100%和100%,LC50值分别为1.03、9.68、3.08和2.87 mg/L。

    • 摘要: The fermentation medium for highly toxic Bacillus sphaericus strain NBIM-311 against Culex quinquefasciatus was optimized through single factor screening and orthogonal experiments. The optimum medium was composed of 20.0 g/L fish meal,15.0 g/L corn steep liquor,60.0 g/L peanut meal,7.5 g/L yeast extract,1.0 g/L Calcium carbonate,0.3 g/L Magnesium sulphate and 0.1 g/L Potassium dihydrophosphate. The pilot fermentation of the strain was successfully carried out in 200 L and 5000 L fermentor,the LC50 of the fermentation broth were 0.963 mg/L and 0.852 mg/L respectively. The LC50 of the pre-pared raw powder obtained from post-treatment of the fermentation broth were 0.0189 mg/L and 0.0223 mg/L respectively.%利用单因素筛选和正交试验,对球形芽孢杆菌(Bacillus sphaericus)NBIM-311高毒菌株的发酵培养基进行优化,得到其最佳培养基配方为鱼粉20.0 g/L,玉米浆15.0 g/L,花生粕60.0 g/L,酵母膏7.5 g/L,碳酸钙1.0 g/L,硫酸镁0.3 g/L,磷酸二氢钾0.1 g/L.通过200 L和5000 L发酵罐分别发酵,发酵液对致倦库蚊致死中浓度分别为0.963 mg/L和0.852 mg/L;发酵液经过后处理所制备的原粉对致倦库蚊致死中浓度分别为0.0189 mg/L和0.0223 mg/L.

    • 摘要: 目的 构建表达蝎毒素AaIT或苏云金杆菌毒素Cyt2Ba的大肠埃希菌,检测重组菌对致倦库蚊和白纹伊蚊II龄幼虫的毒力,并测试不同剂型的增效作用.方法 分别将AaIT和Cyt2Ba编码基因克隆于pET28a(+)质粒上,并将重组质粒分别转化BL21 E.coli获得分别表达AaIT和Cyt2Ba的工程菌株,经IPTG诱导后,用SDS-PAGE和Western-blot检测重组蛋白的表达;测试不同浓度工程菌菌液对于蚊幼虫的杀灭效果;并将有效的工程菌制成干粉剂,检测其干粉末及其配剂对蚊幼虫的杀灭效果.结果 AaIT和Cyt2Ba重组蛋白在大肠埃希菌中成功表达,但AaIT重组蛋白对蚊幼虫不具有毒性.表达Cyt2Ba重组蛋白的工程菌菌液对白纹伊蚊和致倦库蚊幼虫均具有明显的杀灭作用,48 h的LC50分别为3.00×106和1.25×107 cells/mL,其中对白纹伊蚊幼虫的杀灭效果要显著优于对致倦库蚊幼虫的杀灭效果(P<0.001),并且表达Cyt2Ba重组蛋白的工程菌干粉末及其配剂均具有较好的杀蚊幼虫效果.当该工程菌干粉末与干酵母粉、小麦粉和白胡椒粉分别按4:1、1:1和1:4比例制成配剂处理蚊幼虫48 h后,结果显示当质量比为1:1时,配剂杀蚊幼效果显著提高(P=0.044),并且白胡椒粉末配剂效果要显著好于干酵母粉和小麦粉配剂(P=0.002).结论 表达Cyt2Ba的重组大肠埃希菌对蚊幼虫具有较好的杀灭作用,合适配剂的使用可以增强其在蚊虫防制中的效果.%Objective To assess the larvicidal effects of recombinant Escherichia coli expressing scorpion neurotoxin AaIT or Bacillus thuringiensis subsp israelensis (B.t.i) toxin Cyt2Ba against the second instar larvae of Culex pipiensquinquefasciatus and Aedes albopictus and compare different formulations for their larvicidal effects. Methods The AaIT-or Cyt2Ba-coding sequences were cloned into pET28a(+) and the recombinant plasmids were transformed into E. coli BL21(DE3). After induction with IPTG, the recombinant proteins expressed by the recombinant E. coli were detected and identified by SDS-PAGE and Western blotting, respectively. The larvicidal activity of the bacterial suspension was tested at different concentrations against mosquitoes. The effective engineered bacteria were prepared into dry powder with different formulations, and their larvicidal activity was tested. Results AaIT and Cyt2Ba proteins were successfully expressed in E. coli. The recombinant AaIT protein showed no virulence to the mosquito larvae. The suspension of the recombinant E. coli expressing Cyt2Ba protein exhibited a stronger killing effect on Aedes albopictus larvae than on Culex pipiens quinquefasciatus larvae at 48 h (P<0.001) with LC50 of 3.00× 106 cells/mL and 1.25 × 107 cells/mL, respectively. The dry powder of the engineered bacteria formulated with yeast extract, wheat flour or white pepper powder at the mass ratio of 1:1 showed the strongest killing effect on mosquito larvae (P=0.044), and the formulation with white pepper powder produced a stronger killing effect than formulations with yeast extract or wheat flour (P=0.002). Conclusion The B.t.i Cyt2Ba protein expressed in E. coli BL21(DE3) shows a good larvicidal activity against mosquitoes, and appropriate formulations of the engineered bacteria can enhance its efficiency in mosquito control.

    • 摘要: Objective:To investigate the infection status and genotype of symbiotic bacteria (Wolbachia) in common mosquito in Guiyang.Methods:Mosquitoes were collected by using a spoon from 6 districts of Guiyang (Yunyan,Nanming,Huaxi,Xiuwen,Xifeng and Kaiyang) and identified by morphology in the lab.Wolbachia surface protein gene (wsp) was amplified by PCR and sequenced for phylogenetic analysis.Results:There were 3 species mosquito found in the different habitats in the 6 districts,Aedes albopictus,Culex pipens pallens and Armigeres obturbans.The positive rate of wsp ranged from 42.9% to 95%.13 target gene sequences were obtained with sequence length between 572 to 602bp.Based on the phylogenetic tree,these sequences were divided into A and B supergroup respectively.The wolbachia genotype of A albopictus was wAlbA and wAlbB.The Wolbachia genotype of Culex pipens pallens was wPip.The Wolbachia genotype of Armigeres obturbans was wAlbA.Conclusions:Aedes albopictus,Culex pipens pallens and Armigeres obturbans in Guiyang are generally infected with Wolbachia and their genotypes are wAlbA,wAlbB and wPip.%目的:了解贵阳市常见蚊虫种群中沃尔巴克体(Wolbachia)的感染情况及基因型.方法:采用勺舀法采集贵阳市6城区(云岩区、南明区、花溪区、修文县、息烽县和开阳县)的蚊虫并鉴定;选用Wolbachia表面蛋白基因(wsp),利用PCR方法对单只雌蚊进行扩增、测序并做系统发育分析.结果:在贵阳市6城区不同水体采集到的常见蚊种有3种,分别是白纹伊蚊、致倦库蚊和骚扰阿蚊,3种蚊虫种群Wolbachia的核酸阳性率为42.9%~95.0%;获得13条目的基因序列、序列长度为572 ~ 602 bp,系统发育树显示这些序列分别属于A和B两个超群,白纹伊蚊Wolbachia基因型为wAlbA和wAlbB,致倦库蚊Wolbachia基因型为wPip,骚扰阿蚊Wolbachia基因型为wAlbA.结论:贵阳市白纹伊蚊、致倦库蚊和骚扰阿蚊普遍感染Wolbachia,其基因型为wAlbA、wAlbB和wPip.

    • 摘要: In order to study the histological changes of Culex pipiens quinquefascitus larvae infected by this fungus and investigate the mechanism of Pythium guiyangense for killing Cx. quinquefascitus larvae. The larvae of Cx. quinquefasciatus at different infection periods were fixed. Their paraffin sections were made and stained with H. E. method, observed with microscope and photoed with NIS⁃Elements. The midgut of Cx. quinquefasciatus larvae were destructed remarkably, the gap of epithelial cells on midgut increases, cell vacuolation, the plasma membrane damage and cell lysis. It was considered as one of the lethal causes of Cx. quinquefasciatus larvae that P. guiyangense destroyed epithelial cells of midgut.%为了观察灭蚊真菌贵阳腐霉感染致倦库蚊幼虫后的组织形态学变化,探讨贵阳腐霉对致倦库蚊幼虫的致死机理,采用石蜡切片、 H. E.染色和比较组织学方法,以未经贵阳腐霉作用的致倦库蚊幼虫为对照,连续观察用贵阳腐霉感染24~72 h致倦库蚊幼虫的组织学变化。结果发现被贵阳腐霉感染的致倦库蚊幼虫中肠上皮细胞出现细胞间隙增大,随着感染时间的延长,细胞间隙进一步增大并出现空泡化、质膜破损、细胞裂解一系列病理变化。结果表明贵阳腐霉可导致致倦库蚊幼虫中肠上皮细胞的破坏,推测这可能是导致其死亡原因之一。
