    • 摘要: 为探究安徽小麦品种高分子量麦谷蛋白亚基(HMW-GS)组成及品质性状,对97份安徽省主要种植的小麦品种进行了HMW-GS组成及品质性状分析。结果表明,供试材料中共检测出10种HMW-GS亚基类型和19种亚基组合,在Glu-A1位点检测出3种亚基类型,以1亚基(57.73%)和Null亚基(40.21%)为主;在Glu-B1位点共检测出3种亚基类型,以7+8亚基(52.58%)为主;在Glu-D1位点共鉴定出4种亚基类型,优质亚基5+10占比最大,为45.36%;供试品种亚基组合品质得分在4~10分之间,10分的亚基组合1/7+8/5+10和1/17+18/5+10共19份材料(19.59%),8分以下的共44份材料(45.36%)。对供试材料HMW-GS与品质性状进行相关分析发现,Glu-1的三个位点与硬度指数、溶剂保持力和吸水率相关达显著或极显著水平,对溶剂保持力影响表现为Glu-A1>Glu-B1>Glu-D1;不同亚基组合对品质性状的影响存在差异,亚基组合为1/17+18/5+10的品种具有最高的硬度指数、4种溶剂保持力、吸水率、稳定时间和粉质质量指数,具有最低的L*和W,该类品种有烟农19、泰农19、山农17和糯小麦1012;亚基组合为Null/7+9/4+12的品种具有最低的硬度指数、4种溶剂保持力、吸水率、稳定时间和粉质质量指数,具有最高的弱化度、L*和W,该类品种有荃麦725、皖麦52和未来0818。

    • 摘要: 【目的】研究新疆春小麦及CIMMYT引进材料的HMW-GS组成差异,为新疆春小麦杂交选配提供依据。【方法】采用SDS-PAGE电泳技术。分析新疆春小麦192份材料中的HMW-GS的组成及分布频率。【结果】供试材料中共检测出13种HMW-GS等位变异,Glu-A1位点等位变异Null亚基频率最高(64.06%),其次为1亚基(21.35%)和2*亚基(14.58%)。Glu-B1位点等位变异类型表现最丰富,其亚基频率高低顺序为7+8(39.58%)>7(17.19%)>7+9(13.54%)>17+18(11.46%)>22(9.38%)>13+16(7.29%)>6+8(1.04%)>14+15(0.52%);其中7+8、17+18、14+15和13+16亚基为优质亚基,频率为58.85%。Glu-D1位点检测2+12(49.48%)和5+10(50.52%)频率较为接近。在Glu-A1、Glu-B1和Glu-D1位点的优质亚基比例均表现为新疆春小麦品种(系)>引进CIMMYT材料。【结论】全部HMW-GS共形成32种亚基组合,品质评分平均分为7.04分,其中新疆春小麦近年育成的新品种(系)表现出品质评分较高平均分为8.12分,明显高于引进CIMMYT材料品质评分平均为6.09分。评分为10分的材料有25个,评分在8分以上的有90份材料。

    • 摘要: High-molecular-weight glutenin subunits(HMW-GSs) play a critical role in determining the viscoelastic properties of wheat. As the organelle where proteins are stored, the development of protein bodies(PBs) reflects the status of protein synthesis and also affects grain quality to a great extent. In this study, with special materials of four near-isogenic lines in a Yangmai 18 background we created, the effects of Glu-A1 and Glu-D1 loci deletions on the development and morphological properties of the protein body, protein components and dough properties were investigated. The results showed that the deletion of the HMW-GS subunit delayed the development process of the PBs, and slowed the increases of volume and area of PBs from 10 days after anthesis(DAA) onwards. In contrast, the areas of PBs at 25 DAA, the middle or late stage of endosperm development, showed no distinguishable differences among the four lines. Compared to the wild type and single null type in Glu-A1, the ratios of HMW-GSs to low-molecular-weight glutenin subunits(LMW-GSs), glutenin macropolymer(GMP) content, mixograph parameters as well as extension parameters decreased in the single null type in Glu-D1 and double null type in Glu-A1 and Glu-D1, while the ratios of gliadins(Gli)/glutenins(Glu) in those types increased. The absence of Glu-D1 subunits decreased both dough strength and extensibility significantly compared to the Glu-A1 deletion type. These results provide a detailed description of the effect of HMW-GS deletion on PBs, protein traits and dough properties, and contribute to the utilization of Glu-D1 deletion germplasm in weak gluten wheat improvement for use in cookies, cakes and southern steamed bread in China and liquor processing.

    • 摘要: 内麦品种是西南麦区中筋小麦代表品种之一,在四川及周边省份种植面积累计达330万hm2以上.利用SDS-PAGE电泳技术对内麦品种进行高分子量谷蛋白亚基(HMW-GS)组成进行分析,发现内麦品种中与小麦加工品质呈正相关的5+10优质亚基出现频率达85.7%,面包评分达7分.经测定内麦品种面条评分高于同期主栽品种.选育含5+10优质亚基的小麦品种不仅能满足西南地区对中筋面条小麦的市场需求,同时也为提升中国小麦品质作出贡献.

    • 摘要: 高分子量麦谷蛋白亚基(HMW-GS)是决定小麦加工品质的重要因素.高效毛细管电泳技术(HPCE)以其用样少、速度快、精确度高等特点被越来越多地应用到分离鉴定中来.目前小麦 HMW-GS 的 HPCE 研究尚处于起步阶段, 对标准鉴定图谱的研究甚少, 且已有分离体系在连续多次试验中的分离速度及分辨率仍有提升空间.本研究通过SDS-PAGE结合分子标记的方法鉴定了不同小麦品种的HMW-GS, 以"中国春"小麦为标样确立了HPCE高效分离体系, 体系条件为75 mmol L–1IDA+0.05% HPMC+15% ACN, pH 2.5, 电泳参数为毛细管内径25 μm, PDA检测波长200 nm, 分离电压20 kV, 运行温度30°C.通过混合进样方式对不同类型亚基进行HPCE分析, 建立了18个亚基的标准图谱, 亚基迁移顺序为 1Dy12→1Dy10→1By9→1By8→1By18→1By16→1By20→1Bx17→1Bx20→1Bx13→1Bx6→1Bx7→1Ax2*→1Ax1→1Dx5→1Dx4→1Dx3→1Dx2, 标准出峰时间依次为9.39、9.69、10.30、11.70、11.89、12.09、12.22、12.36、12.62、12.83、13.08、13.18、13.50、13.73、14.04、14.24、14.46和14.73 min, RSD<0.2%.以1Bx17为分界线, 9.39 min到12.36 min为y型亚基区域, 12.36 min到14.76 min为x型亚基区域.结合亚基迁移顺序、标准出峰时间及图谱特点可对小麦相关 HMW-GS 进行 HPCE 快速鉴定.本研究获得的分离体系及鉴定图谱可用于小麦HMW-GS定性分析和种质资源筛选.%High molecular weight glutenin subunit (HMW-GS) is important for processing quality of wheat. High performance capillary electrophoresis (HPCE) is increasingly used in the work of separation and identification due to its advantages of small sample, rapidness and high precision. However, this technique has been seldom used in wheat HMW-GS study and its separation system needs improvement in analytical speed and discernibility. On the basis of HMW-GS identification with SDS-PAGE and molecular markers, an high-efficiency separation system of HPCE was set up using Chinese Spring as the standard. The system components (pH 2.5) were 75 mmol L–1IDA, 0.05% HPMC, and 15% ACN. The electrophoresis parameters were 25 μm of inner diameter of the capillary, 200 nm of detection wavelength, 20 kV of separation voltage, and 30°C of operating temperature. Using mixed injection method, the standard spectrums were obtained for 18 subunits. Their migration order was 1Dy12→1Dy10→1By9→1By8→1By18→1By16→1By20→1Bx17→1Bx20→1Bx13→1Bx6→1Bx7→1Ax2*→1Ax1→1Dx5→1Dx4→1Dx3→1Dx2, and standard peak time of these subunits was 9.39, 9.69, 10.30, 11.70, 11.89, 12.09, 12.22, 12.36, 12.62, 12.83, 13.08, 13.18, 13.50, 13.73, 14.04, 14.24, 14.46, and 14.73 min, respectively. The relative standard deviation was smaller than 0.2%. The y-type and x-type subunits appeared in the phases of 9.39–12.36 min and 12.36–14.76 min, respectively, between which there was 1Bx17 as the boundary. These results indicate that the HPCE separation system is applicable in rapid identification of HMW-GS in wheat germplasm resources when we simultaneously consider migration order, standard peak time, and HPCE spectrum.

    • 摘要: This experiment detected and analyzed 11 foreign wheat cultivars resources of high molecular weight glutenin subunits (HMW-GS) composition,Glu-1 locus mutation and the frequency by conventional SDS-PAGE technology.The results showed that the Glu-A1 loci encoding HMW-GS had two types,2* and null.Glu-B1 loci encoding HMW-GS had four types,namely 7 + 9,6 + 8,7 + 18 and 6.8 + 20y,Glu-D1 loci encoding HMW-GS had 4 + 12,2 + 6.5,2 + 10,2 + 9,6.5 and 6 + 13 six types.The occurrence frequency of 2* subunit of Glu-A1 site,6 + 8 subunit of Glu-B1 site and 4 + 12 subunit of Glu-D1 site were higher.%以11份国外小麦品种资源为材料,利用聚丙烯酰胺凝胶电泳(SDS-PAGE)技术,对小麦材料高分子量谷蛋白亚基(HMW-GS)的组成、Glu-1位点变异类型及亚基出现的频率进行检测分析.结果表明,小麦品种的Glu-A1位点编码的HMW-GS有2种类型,即2*和Null;Glu-B1位点编码的HMW-GS有4种类型,即7+9,6+8,7+18和6.8+ 20y;Glu-D1位点编码的HMW-GS有6种类型,即4+12,2+6.5,2+10,2+9,6.5和6+13.其中,Glu-Al位点上的2*亚基、Glu-B1位点上的6+8亚基、Glu-D1位点上的4+12亚基出现频率最高.

    • 摘要: To improve the efficiency of high quality wheat breeding program,clarify the composition of HMW-GS and Gliadin in the early generation,296 lines from the wheat maize cross were analyzed useing SDS-PAGE meth-od in this experiment,along with this Gliadin were analyzed useing A-PAGE method.A statistics showed that female parent Heng 09-6324 carried null,7 +9,2 +12 subunits,and were 1 BL /1 RS translocation lines with Sec-1 charac-teristic band.Male parent Shiluan 02-1 carried 1 ,7 +9,5 +10 subunits,and were non 1 BL /1 RS translocation.Six types of HMW-GS all originated from parents were identified from 296 DH lines.The frequency of subunits 5 +10 which positively related to bread baking quality was 35.13%.A statistics showed that in the A-PAGE the type of translocation of the DH lines accounted for 54.05%,and the percent of lines with subunits 1 and 5 +10,but non Sec-1 translocation were 15.20%,these lines could be used as new germplasm for quality breeding.The paper indi-cated that that the method of half-seed SDS-PAGE and A-PAGE which could be used to identify and select excellent plants in early generation breeding effectively.%为了提高优质小麦的育种效率,在早代明确高分子量谷蛋白亚基和醇溶蛋白的组成,增加育种的针对性,以1个小麦杂交组合的296个 DH 株系群体为试材,采用半粒 SDS-PAGE 法检测麦谷蛋白,且用 A-PAGE 检测了醇溶蛋白。结果表明:母本衡09-6324的 HMW-GS 构成为 null、7+9、2+12,是有 Sec-1特征带的1 BL /1 RS 易位系品种,父本师栾02-1亚基组成1、7+9、5+10,是非1 BL /1 RS 易位系品种。296个 DH 株系共鉴定出6种不同的 HMW-GS 类型,均为父母本表达亚基类型的再分配,无变异类型。与面包加工品质呈正相关的5+10亚基出现频率为35.13%。DH 株系材料中1 BL /1 RS 易位系类型的占54.05%,1、5+10优质亚基同时出现,且没有 Sec-1表达的占15.20%,这些株系可作为新的面包小麦种质资源使用。采用半粒 SDS-PAGE 和 A-PAGE 法对育种早代的优异单株进行有效鉴定和选择,可以提高优质小麦育种效率。

    • 摘要: 以3个不同小麦品种中国春、宁春4号和宁春42号为材料,在N、P2O5、K2O不同营养水平下进行栽培(盆栽)试验,利用SDS-PAGE电泳等分析技术,研究肥力营养对麦谷蛋白和HMW-GS亚基动态形成的影响.结果表明:(1)在不同水平施肥处理下各品种开花后谷蛋白形成时间不尽相同,但开花后麦谷蛋白积累变化均呈"低-高-低"总趋势,供试3个品种随籽粒发育成熟谷蛋白含量逐渐增多,大约在开花后25d总含量达最大积累,之后稍有下降;(2)在不同施肥处理水平下3个品种高分子量谷蛋白亚基花后5d开始至15d全部形成,且品种不同亚基形成时间不同,同一品种内亚基形成顺序不同,以Glu-D lx和Glu-B1x位点编码的亚基最早出现,Glu-A1编码的亚基形成最晚;(3)氮、磷、钾肥影响供试品种宁春4号、宁春42号高分子量谷蛋白亚基的组成.

    • 摘要: 用200 Gy的60Co-γ射线辐射强筋小麦品种郑9023,对M2代918株的产量和品质性状进行遗传变异分析和SDS-PAGE电泳鉴定.结果表明,以超过M2代群体均值±1.96×标准差确定变异单株,单株产量、谷蛋白大聚合体(GMP)含量和谷蛋白大聚合体/蛋白质(GMWPr)仅产生了正向变异,变异单株分别为43个、26个和25个;蛋白质含量42个单株发生正向变异,7个单株发生负向变异;湿面筋含量40个单株发生正向变异,10个单株发生负向变异;硬度15个单株发生正向变异,45个单株发生负向变异;沉降值25个单株发生正向变异,24个单株发生负向变异;PPO活性21个单株发生正向变异,16个单株发生负向变异.以上各性状的变异系数在6.3%~50.6%之间.相关分析表明,GMP含量与蛋白质含量、湿面筋含量、沉降值呈极显著正相关,GMP含量与单株产量呈极显著负相关.PPO活性与产量和品质性状皆无显著相关性.经SDS-PAGE电泳鉴定,有6个高分子量谷蛋白亚基(HMW-GS)变异单株,高分子量谷蛋白亚基从郑9023的N/7 +8/2+12突变为1/7+ 8/2+ 12,品质得分增加2分.

    • 摘要: 应用SDS-PAGE方法对24份外引品种和31份品种(系)的HMW-GS组成与分布进行分析。结果表明,HMW-GS分别具有17种带型组合;3个位点共发现14个等位基因。Glu-A1、Glu-B1和Glu-D1分别有3、7和4个变异位点;亚基Null、1、7+9、7+8、2+12和5+10是主要的HWM-GS类型,频率分别为61.8%、36.4%、20.0%、45.5%、56.4%和36.4%。
