    • 摘要: 目的 评价前列腺干细胞抗原(prostate stem cell antigen,PSCA)基因rs2976392多态位点与中国人群胃癌易感性的关联.方法 通过计算机检索中国知网、万方数据库、维普数据库、NCBI、Embase、Web of Science等数据库获取关于胃癌与PSCA基因rs2976392多态位点相关的文献,检索时间为自建库起至2020年3月22日.对纳入文献进行质量评价后,使用RevMan 5.3和Stata 12.0软件进行Meta分析.结果 最终纳入13篇文献共14项病例对照研究,包含6618例胃癌患者和6952例对照者.Meta分析显示,等位基因模型(A vs G)、显性基因模型(AA+GA vs GG)和1个共显性基因模型(AA vs GG)均显示PSCA基因rs2976392多态位点与中国人群胃癌发病风险增加有关(OR=1.28,95%CI:1.08~1.52,P=0.004;OR=1.35,95%CI:1.27~1.45,P0.05).结论 PSCA基因rs2976392多态位点与中国人群胃癌发病风险升高有关,且以纯合AA基因型汉族人群的胃癌发病风险最高.

    • 摘要: 嵌合抗原受体修饰T细胞(CAR-T)是近年来免疫治疗的热点,在血液系统疾病的治疗中取得了显著的临床疗效.目前,CAR-T在前列腺癌(PCa)治疗的研究中也取得了较大进展,其中以前列腺特异性膜抗原(PSMA)和前列腺干细胞抗原(PSCA)为靶点的CAR-T治疗进展较快,其可显著抑制肿瘤细胞生长,一定程度地延缓疾病进展.本文综述了前列腺肿瘤相关抗原(TAA)在前列腺癌CAR-T治疗中的最新研究进展,并对CAR-T在前列腺癌治疗中的挑战予以简要探讨,以期为临床提供参考.

    • 摘要: 目的:探究外周血中的前列腺特异性抗原(PSA)以及前列腺干细胞抗原(PSCA)单独或进行联合检测在前列腺癌临床诊断中的应用价值.方法:选取本院2016-08 ~2018-08本院确诊前列腺增生患者50例建立BPH组、前列腺癌患者50例建立PCA组以及同期进行健康体检者50例建立对照组.通过酶联免疫吸附试验检测三组研究对象血清PSA以及PSCA含量,通过受试者工作特征曲线(ROC)评价PSA以及PSCA单独或进行联合检测对于前列腺癌诊断的价值.结果:相比于对照组,BPH组以及PCA组的血清PSA以及PSCA含量均有明显的升高(P<0.01);PCA组相比于BPH组的PSA以及PSCA含量均有明显的升高(P<0.01).对照组与BPH组中,血清PSA与PSCA的联合检测ROC曲线下面积相比于两指标的单独检测显著更高;对照组与PCA组、BPH组与PCA组中,血清PSA与PSCA的联合检测ROC曲线下面积相比于单独PSA检测均显著更高,但联合检测与单独PSCA检测的对比未见显著差异;三组检测均显示联合检测的诊断价值更高.结论:在健康人与前列腺疾病的患者外周血PSA以及PSCA的表达上存在有显著差异,提示外周血PSA以及PSCA的检测具有诊断BPH与PCA的价值.且ROC曲线分析提示,对外周血PSA与PSCA的单独对BPH与PCA上具有一定的诊断价值,联合检测的效果更佳.

    • 摘要: 扩增前列腺干细胞抗原(PSCA)及其特异单链抗体PaFv的编码基因,分别克隆到pET-32a和pDAP2/S载体中,转化大肠杆菌感受态细胞.融合蛋白TrxA-PSCA和PaFv-AP诱导表达后,SDS-PAGE和Western blot分析其表达情况,碱性磷酸酶(AP)显色反应和ELISA检测蛋白的活性.结果 显示构建的原核表达体系成功实现TrxA-PSCA和PaFv-AP融合蛋白的可溶性表达,而且PaFv-AP具有与PSCA抗原结合的抗体活性以及AP活性.

    • 摘要: 目的 探讨前列腺增生患者与前列腺癌患者外周血中前列腺干细胞抗原mRNA表达水平的差异.方法 选取我院2017年1月~12月期间收治的30例良性前列腺增生患者及30例前列腺癌患者为研究对象.采用RT-PCR法测定前列腺增生与前列腺癌患者外周血前列腺干细胞抗原(PSCA)mRNA表达情况.结果 前列腺癌组受试者外周血PSCA mRNA阳性表达率明显高于前列腺增生组(P<0.05).低分化组前列腺癌患者外周血PSCA mRNA阳性表达率明显高于中、高分化组(P<0.05),中分化组明显高于高分化组(P<0.05).结论 前列腺癌患者外周血PSCA mRNA表达水平明显高于良性前列腺增生患者,且前列腺癌患者外周血PSCA mRNA表达水平与肿瘤分化程度相关,呈负相关系.

    • 摘要: 目的 系统评价前列腺干细胞抗原(PSCA) rs2294008位点多态性与胃癌癌前病变是否存在相关性.方法 应用计算机检索电子数据库PubMed 、SinoMed、CNKI、万方、VIP,按纳入标准搜索含有研究PSCA rs2294008多态性与胃癌癌前病变相关性的文章,并对所纳入文献进行Meta分析.结果 共5篇文章纳入研究,病例组2 310例,对照组3 240例,研究表明,PSCAra2294008等位基因模型、加性模型、共显性模型、显性模型与胃癌癌前病变的易感性有关,差异有统计学意义(T vs C:OR=1.18,95% CI:1.09~1.28,P<0.0001;TTvs CC:OR=1.46,95% CI:1.08 ~1.97,P =0.01;CT vs CC:OR=1.25,95% CI:1.09 ~1.42,P =0.0009;CT +TT vs CC:OR=1.31,95% CI:1.16~1.49,P<0.0001),隐性模型与胃癌癌前病变的易感性有关,但差异无统计学意义(TT vsCT+CC:OR=1.24,95% CI:0.93~1.66,P=0.15).PSCA突变等位基因T增加了萎缩性胃炎的易感性,差异有统计学意义(TvsC:OR=1.24,95% CI:1.12~1.38,I2 =24,P<0.0001;TTvs CC:OR=1.60,95% CI:1.28~2.01,I2 =3,P<0.0001;CTvs CC:OR=1.42,95% CI:1.17~1.72,I2 =0,P=0.0004;CT+TTvs CC:OR=1.47,95% CI:1.22 ~1.77,I2 =0,P <0.0001;TTvs CC:OR=1.24,95% CI:1.04~1.47,I2 =47,P=0.02).结论 PSCA rs2294008位点多态性与胃癌癌前病变有相关性.%Objective To systematically evaluate the relationship between prostate stem cell antigen (PSCA) rs2294008 polymorphisms and gastric cancer precancerous lesions.Methods Articles were searched according to the included standards that contained PSCA rs2294008 polymorphisms and gastric cancer precancerous lesions from PubMed,SinoMed,CNKI,WanFang and VIP.Results A total of 5 articles were included in the study,involved 5 550 individuals (2 310 in the case group,3 240 in the control group).Meta-analysis showed that the allele model,additive model,codominant model,dominant model of PSCA rs2294008 were related with the susceptibility of gastric cancer precancerous lesions.And the differences were statistically significant (P < 0.05).The recessive model was associated with susceptibility of gastric precancerous lesions,but the difference was not statistically significant (P > 0.05).PSCA risk allele T increased the susceptibility of atrophic gastritis.The difference was statistically significant (P < 0.05).Conclusion PSCA rs2294008 may have an impact on the risk of susceptibility of gastric precancerous lesions.

    • 摘要: Objective:To investigate the expression of PSCA and TBX2 in hepatocellular carcinoma (HCC)speci-mens. Methods:The expression of PSCA and TBX2 was detected by immunohistochemistry method in 64 formalin-fixed,paraffin-embedded HCC and and corresponding paired adjacent noncancerous tissues. Results:The expression of PSCA and TBX2 in HCC were increased remarkably (73% and 75% respectively),compared with those in adja-cent noncancerous tissues,respectively (P<0. 05). The expression of PSCA was localized in the cytoplasm and TBX2 in the cytoplasm and cytoplasmuclei of hepatic cell. The expression of PSCA and TBX2 in HCC was not correla-ted with gender and age ,respectively (P>0. 05),but correlated with stage ,grade and metastasis ,respectively (P<0. 05). Conclusion:The abnormal expression of PSCA and TBX2 was related with carcinogenesis of HCC. PSCA and TBX2 might be candidate targets for the therapy of HCC.%目的:初步研究肝细胞癌组织中前列腺干细胞抗原(prostate stem cell antigen,PSCA)和T-box转录因子(T-box transcription factors,TBX2)的表达情况及临床意义.方法:收集64例甲醛固定石蜡包埋的肝细胞癌和癌旁组织,利用SP免疫组化方法检测组织中PSCA和TBX2的表达情况.结果:在肝细胞癌组织中PSCA和TBX2的表达率分别为73%和75%,均高于癌旁组织中的表达(P<0. 05).PSCA蛋白表达位于肝细胞的胞浆中而TBX2蛋白表达位于肝细胞胞浆或者胞核中.肝细胞癌组织中PSCA和TBX2的表达与性别、年龄无关(P>0. 05),但是分别与临床分期、病理分级和转移有关(P<0. 05).结论:PSCA和TBX2的异常表 达与肝细胞癌变有关,PSCA和TBX2可能会成为肝细胞癌的治疗靶点.

    • 摘要: 目的 探讨血清前列腺干细胞抗原(PSCA)、前列腺特异性抗原(PSA)联合检测在前列腺癌骨转移诊断中的意义.方法 经前列腺穿刺活检初次病理确诊的前列腺癌患者86例,根据放射性核素骨显像结果分为骨转移(43例)与非骨转移(43例)两组,以骨转移3级及以上划分为进展性(19例)和局限性骨转移(24例).采用化学发光免疫染色法对血清PSA进行检测,利用酶联免疫吸附试验法测定PSCA水平,采用受试者工作特征曲线(ROC)评估两种标志物诊断前列腺癌骨转移的价值.结果 骨转移组的血清PSCA、PSA含量明显高于非骨转移组(P<0.05).与局限性骨转移组比较,进展性骨转移组的PSCA和PSA水平均有明显升高(P<0.05).骨转移患者的PSCA、PSA、Gleason评分与骨转移数目呈正相关(r=0.730 1,P<0.01;r=0.683 8,P<0.01;r=0.422 0,P<0.01),而临床T分期与骨转移数目无明显相关性(r=0.251 4,P>0.05).PSCA、PSA、PSCA+ PSA诊断前列腺癌骨转移的ROC曲线下面积(AUC)分别为0.770、0.840、0.886;其灵敏度分别为60.47%、79.07%、81.40%;其特异度分别为88.37%、83.72%、95.35%;其正确诊断率分别为74.42%、81.39%、82.56%;血清PSCA、PSA诊断前列腺癌骨转移的临界值分别为119.31 ng/mL、43.59ng/mL.结论 血清PSCA、PSA诊断前列腺癌骨转移的价值相当,联合检测能提高前列腺癌骨转移的诊断效能,有利于骨转移的早期诊断.

    • 摘要: 目的 探讨外周血中前列腺特异性抗原(PSA)和前列腺干细胞抗原(PSCA)单独或联合检测对于前列腺癌的临床诊断价值.方法 选取100名健康者(非患者组)、91例前列腺增生患者(BPH组)和84例前列腺癌患者(PCA组)作为研究对象,采用酶联免疫吸附试验检测各组血清中PSA和PSCA的含量,并采用受试者工作特征(ROC)曲线评价单独或联合检测PSA和PSCA对前列腺癌的诊断价值.结果 BPH组和PCA组血清中PSA和PSCA含量显著高于非患者组(P<0.01),PCA组中PSA和PSCA含量显著高于BPH组(P<0.01).PSA、PSCA单独或联合检测区分非患者和BPH患者的ROC曲线下面积分别为0.690(95%CI为0.615~0.766),0.700(95%CI为0.673~0.814)和0.898(95%CI为0.854~0.943),表明两指标单独或联合检测对于BPH患者具有一定的诊断意义;区分非患者和PCA患者的曲线下面积分别为0.867(95%CI为0.813~0.921),0.904(95%CI为0.858~0.951)和0.973(95%CI为0.944~1.000),表明PSA、PSCA单独或联合检测对于PCA诊断具有较高的价值;区分BPH患者和PCA患者的曲线下面积分别为0.687(95%CI为0.608~0.766),0.818(95%CI为0.752~0.884)和0.862(95%CI为0.803~0.920),表明在区分BPH和PCA的诊断中,二者单独检测均具有一定价值,且联合检测具有较高的临床诊断意义.结论 血清PSA和PSCA单独或联合检测对于BPH和PCA均具有临床诊断价值,并且联合检测的诊断价值更高.%Objective To investigate the clinical value of prostate-specific antigen (PSA) and prostate stem cell antigen (PSCA) in peripheral blood alone or in combination for the clinical diagnosis of prostate cancer.Methods 100 healthy people (non-patient group), 91 patients with benign prostatic hyperplasia (BPH group) and 84 patients with prostate cancer (PCA group) were selected.The levels of PSA and PSCA were measured by enzyme-linked immunosorbent assay.The diagnostic value of PSA and PSCA alone or in combination for the diagnosis of prostate cancer was evaluated using the receiver operating characteristics (ROC) curve.Results The levels of PSA and PSCA in BPH group and PCA group were significantly higher than those in non-patients group (P<0.01), and the levels of PSA and PSCA in PCA group were significantly higher than those in BPH group (P<0.01).The area under the ROC curve for PSA and PSCA alone or in combination to distinguish between non-patients and BPH patients were 0.690(95%CI 0.615-0.766), 0.700(95%CI 0.673-0.814) and 0.898(95%CI 0.854-0.954) respectively,indicating that the two indicators alone or in combination had a certain diagnostic significance for BPH patients;the area to distinguish between non-patients and PCA patients were 0.867(95%CI 0.813-0.921), 0.904(95%CI 0.858-0.951) and 0.973(95%CI 0.944-1.000) respectively, which indicated that the detection of PSA, PSCA alone or in combination had a high diagnostic significance for PCA;the area to distinguish between BPH patients and PCA patients were 0.687(95%CI 0.608-0.766), 0.818(95%CI 0.752-0.884) and 0.862(95%CI 0.803-0.920), respectively, indicating that for the distinction between BPH and PCA in the diagnosis, PSA or PSCA alone had a certain diagnostic significance, and the combined detection had a higher clinical significance.Conclusion Both serum PSA and PSCA alone or in combination have significant clinical diagnosis values for both BPH and PCA, and the diagnostic value of combined detection is higher.

    • 摘要: 目的 探讨前列腺穿刺患者的前列腺干细胞抗原(PSCA)基因单核苷酸多态性(SNP)与前列腺癌(PCa)的关系.方法 检测296例经直肠前列腺穿刺活检患者的PSCA rs1045531的SNP,比较PCa和良性前列腺增生(BPH)患者间rs1045531基因型分布频率的差异,并分析其与PCa风险和前列腺特异性抗原(PSA)等参数的关系.结果 PCa组(n=138)与BPH组(n=158)患者的PSCA rs1045531基因型分布频率差异有统计学意义(P=0.000).与CC型比较,AC型显著增加PCa的患病风险[比值比(OR)=2.612,95%可信区间(CI):1.607~4.245,P=0.000].在所有穿刺患者中,AC型患者PSA明显高于CC型[(53.49±29.24) ng/ml比(22.42±18.74) ng/ml,P=0.001];在PCa患者中,AC型患者PSA也明显高于CC型患者[(69.89±43.00) ng/ml比(24.34±11.73) ng/ml,P=0.025].rs1045531 SNP与Gleason评分密切相关(x2=10.370,P=0.006).结论 PSCA rs1045531 SNP可用于前列腺穿刺患者的PCa风险预测和危险评估.%Objective To investigate the association between prostate stem cell antigen (PSCA) gene single nucleotide acid polymorphism (SNP) and prostate cancer (PCa) in patients undergone prostate biopsy.Methods DNA from 296 patients undergoing prostate biopsy was typed for PSCA rs1045531 SNP.The frequency of rs1045531 polymorphism between PCa patients and benign prostatic hyperplasia (BPH) patients were compared.The associations between its polymorphism with the risk of PCa and prostate specific antigen (PSA) were analyzed.Results Frequency of PSCA rs1045531 genotypes were significant difference between PCa and BPH patients (P =0.000).Patients with AC genotype had a significantly higher risk for developing PCa compared to patients with CC genotype [odds ratio (OR) =2.612,95% confidence interval (CI):1.607-4.245,P =0.000].The PSA in patients with AC genotype was significantly higher than that in patients with CC.There is association between rs1045531 SNP with Gleason score (x2 =10.370,P =0.006).Conclusion PSCA rs1045531 SNP can be used in evaluating the risk and prognosis of PCa in patients undergoing prostate biopsy.
