The aim was to optimize the extraction process of rice bran soluble dietary fiber (RSDF) by ultrasonic assisted mixed enzyme methods.These factors which influence the extraction efficiency were discussed: the added amount of enzyme, solid to liquid ratio, ultrasonic time and power, water bath temperature and extraction time.The single factor and CCD of response surface analysis showed that the optimum extraction conditions were: papain amount 6%, hydrolysis of 1 h at 60℃ for enzyme inactivation,then adding xylanase dosage 1.5%, solid to liquid ratio of 1∶25 g/ml,ultrasonic time (600 W) of 5 min at 55℃.Under this conditions, the yield of RSDF was up to 25.10%.%对超声辅助混合酶法提取米糠水溶性膳食纤维(RSDF)的提取工艺进行了研究.考察了酶及其添加量、料液比、超声功率和超声时间等对RSDF得率的影响.经过单因素试验和central composite design中心复合响应面分析法,确定了RSDF的最佳提取工艺为:添加6%的木瓜蛋白酶,60℃水解1 h并灭酶后,再添加1.5%木聚糖酶、料液比1∶25 g/ml,55℃水浴超声(600 W)5 min.在此条件下,米糠水溶性膳食纤维得率最高为25.10%.
