中药复方加水解酪蛋白肽粉功效及遗传毒性研究

         

摘要

目的:考察天麻、葛根和罗布麻叶复方加水解酪蛋白肽(casein hydrolysate,CH)粉的降血压功效及遗传毒性。方法分别采用SD大鼠和原发性高血压大鼠(SHR),考察中药复方、CH及两者配伍组方(中药+CH)对动物收缩压、舒张压和心率的影响;以小鼠精子畸形试验、微核试验和Ames试验评价样品的遗传毒性。结果中药+CH的降压作用较中药或CH单用均显著提高(P<0.05),各组样品对心率无显著影响(P>0.05);中药复方组、CH组及两者配伍组精子畸形率和微核发生率与相应试验对照组差异无统计学意义(P>0.05),Ames试验结果均为阴性。结论天麻、葛根和罗布麻叶复方加入CH可显著增强其降血压作用,无遗传毒性。%Objective To observe the antihypertensive effect and genotoxicity of TCM compound composed of Gastrodiae rhizoma, Puerariae lobatae radix and Apocyni veneti folium combined with casein hydrolysate (CH). Methods SD rats and SHR rats were used in the function experiments, the effects of TCM compound, CH and combination of TCM compound and CH (TCM+CH) on systolic pressure (SP), diastolic pressure (DP), and heart rate (HR) were estimated after administration by gavage. The genetoxicity was evaluated by mice sperm abnormality test, micronucleus test and salmonella typhimurium/mammals xicrosomal enzyme test (Ames test). Results The antihypertensive effect of TCM+CH was higher than that of TCM compound or CH administered alone (P<0.05). All the samples showed no significant influence on heart rate. There were no significant difference between the sperm abnormality rate and micronucleus rate of TCM Compound, CH or TCM+CH groups and control group. The results of Ames test were all negative. Conclusion The antihypertensive effect of TCM+CH is higher that of TCM used alone. TCM+CH also has no genotoxicity.
