Objective To investigate the expression of VEGF-D and HMGB1 in gastric carcinoma tissue and their correlation.Methods 87 cases of gastric cancer tissues and paired adjacent tissues and 20 normal gastric tissue specimens in our hospital from January2015 to December 2015 were selected.The expression levels of vascular endothelial growth factor D (VEGF-D) and high mobility group protein B1 (HMGB1) in different samples were detected by immunohistochemical staining, and the correlation between VEGF-D and HMGB1 was analyzed.HMGB1 recombinant plasmid was transfected into human gastric cancer cell line BGC-823, and the expression level of mRNA VEGF-D analyzed by reverse transcription PCR.Results No staining positive cells were found in normal gastric mucosa.48 cases of VEGF-D positive samples (55.17%) and 63 cases of HMGB1 positive samples (72.41%) were detected in 87 cases of gastric carcinoma tissue, while only 22 cases of VEGF-D positive samples (25.29%) and 15 cases of HMGB1 positive samples (17.24%) were detected in 87 cases of adjacent tissue.The positive rates of VEGF-D and HMGB1 in gastric carcinoma were significantly higher than those in adjacent tissues (P<0.05) and normal gastric tissue (P<0.05), and the positive expression of VEGF-D showed positive correlation with HMGB1 (r=0.482, P=0.001).The expression levels of mRNA of HMGB1 and VEGF-D in transfected BGC-823 cells were significantly increased compared with control group, and the differences were statistically significant (P<0.05).Conclusion The expressions of HMGB1 and VEGF were significantly increased in gastric cancer tissue, and they were positively correlated.HMGB1 can promote the transcription of VEGF-D in vitro experiment, but the mechanism remains to be confirmed in further study.%目的 探讨胃癌组织中高迁移率族蛋白B1(HMGB1)、血管内皮生长因子D(VEGF-D)的表达及其相关性,为临床诊断和治疗提供科学依据.方法 选择2015年1月至2015年12月手术切除的87例胃癌组织及其配对的癌旁组织标本为研究对象,另选取20例正常胃组织标本为对照组.采用免疫组织化学染色检测HMGB1和VEGF-D的表达水平,并分析两者的相关性.用HMGB1重组的质粒转染人胃癌细胞BGC-823,用逆转录PCR的方法分析VEGF-D mRNA的表达水平变化.结果 正常胃黏膜中未发现染色阳性细胞,87例胃癌组织共检出VEGF-D阳性标本48例,阳性率为55.17%,检出HMGB1阳性标本63例,阳性率72.41%;87例癌旁组织共检出VEGF-D阳性标本22例,阳性率为25.29%,检出HMGB1阳性标本15例,阳性率为17.24%;胃癌组织中VEGF-D和HMGB1阳性率均显著高于癌旁组织(P<0.05)和正常胃组织(均P<0.05),HMGB1阳性与VEGF-D阳性呈正相关(r=0.482,P=0.001);转染后BGC-823细胞中HMGB1和VEGF-D mRNA表达水平显著增加,差异均有统计学意义(均P<0.05).结论 胃癌组织中HMGB1和VEGF-D蛋白表达水平均显著增加,并且两者呈正相关,体外研究证实HMGB1具有促进VEGF-D基因转录的作用,两者在胃癌发展中的机制还有待进一步研究确认.
