通过形态学观察并结合ITS序列鉴定的方法对真菌菌株产黄青霉AS3.521进行确认,并利用其对根皮素进行转化研究.在28℃、160 r/min与根皮素共同培养3天后,利用水饱和正丁醇溶液萃取发酵液获得转化产物,通过多种柱色谱对转化产物进行分离,运用核磁共振波谱技术(1H-NMR、13C-NMR)结合文献数据鉴定转化产物的结构.结果显示菌株产黄青霉AS3.521可以对根皮素进行转化,且获得了3个小分子转化产物,结构分别为对羟基苯丙酸、对羟基苯甲酸、茴香酸,并推测了转化路径,为化合物合成提供了依据.%Biotransformation of phloretin by Penicillium chrysogenum AS 3.521 was performed and the strain of Penicillium chrysogenum AS3.521 was identified by morphological observation and phylogenetic analysis of the internal transcribed spacer (ITS) gene sequence. The conversion products were obtained in fermentation under the condition of 28 ℃, 160 r/min, then extracted by n-BuOH and separated on macroporous resin, silica gel, octadecyl bonded silica gel(ODS) and preparative high performance liquid chromatography(PHPLC) various column chromatography. The structures of transformation products were elucidated by nuclear magnetic resonance spectroscopy(1H-NMR, 13C-NMR) and compared with literatures. The results showed that the strain was not susceptible through long-term subculture, and still resistant as Penicillium chrysogenum. Three structures were identified as 3-(4-hydroxyphenyl) propanoic acid, p-Hydroxybenzoic acid, anise acid respectively and the conversion path was conjectured, which provided the basis for biosynthesis of compounds.
