苯乳酸高产乳酸菌的分离鉴定及其生长特性的研究

             

摘要

A lactic acid bacterium with high phenyllactic acid (PLA) production was isolated from pickles by dissolving calcium circle tests and agar column diffusion analyses;its identification and growth characteristics were also studied.The best strain among 39 strains with inhibition zone produced 91.5mg/L PLA within 72h.It was identified as Lactobacillus plantarum based on morphological characteristics,physiology and biochemistry tests and 16S rDNA sequence analysis.The optimum initial pH of the medium and culture temperature was 6.0 and 30℃.The stationary stage was reached after 16hrs.cultivation,and PLA production became stable after culturing for 60h.%通过溶钙圈法和琼脂柱扩散法从泡菜中分离到了一株苯乳酸高产乳酸菌,并对其进行了种属鉴定及其生长特性的初步研究.结果表明,在39株具有抑菌效果的菌株中,苯乳酸最高产量为91.5mg/L,经菌落形态、细胞形态、生理生化特性和16s rDNA鉴定表明,该菌株为植物乳杆菌(Lactobacillus plantarum),其最佳生长初始pH为6.0,最佳生长温度为30℃,培养16h即达到稳定期,60h时苯乳酸产量趋于稳定.
