    • 摘要: In the almost seven decades since the initial discovery of the lysosome as an organelle,our understanding of the role of lysosomes has greatly evolved.We now know lysosomal function encompasses far more than its traditionally described role as the cell’s“garbage disposal”,referring to its well-established catabolic function.Lysosomes are integral to maintaining cellular health and viability,and they act as a major signaling hub within the cell(Ballabio and Bonifacino,2020).

    • 摘要: Toothaches have received widespread attention and commentary dating back to ancient times,appearing in Egyptian texts from over 3000 years ago and the writings of Hippocrates,Aristotle,and Galen.Scientific studies of the innervation of dental pulp and dentin date back to at least the 1800’s,yet our understanding of the basis for tooth pain is still limited.Standard treatments for toothache continue to be drastic and irreversible,including removal of the dental pulp or extraction of the tooth.While the trigeminal sensory innervation of the tooth pulp is commonly the origin of toothache pain,it is also indispensable for normal function and physiology,and should be retained as an integral part of the repaired tooth.Repair or regeneration of the dental pulp and its innervation could represent the next step in restorative dentistry,but a better understanding of the roles of the tooth’s sensory nerve supply and the mechanisms underlying its development and repair is needed.

    • 摘要: Spinal cord injury(SCI),either from trauma or degenerative changes,can res ult in severe disability and impaired quality of life.Understanding the cellular processes and molecular mechanisms that underlie SCI is imperative to identifying molecular targets for potential therapy.Recent studies have shown that non-coding RNAs,including both long non-coding RNAs(lncRNAs)and circular RNAs(circRNAs),regulate various cellular processes in SCI.In this review,we will describe the changes in lncRNA and circRNA expression that occur after SCI and how these changes may be related to SCI progression.Current evidence for the roles of lncRNAs and circRNAs in neuronal cell death and glial cell activation will also be reviewed.Finally,the possibility that lncRNAs and circRNAs are novel modulato rs of SCI pathogenesis will be discussed.

    • 摘要: Various time-frequency(T-F)masks are being applied to sound source localization tasks.Moreover,deep learning has dramatically advanced T-F mask estimation.However,existing masks are usually designed for speech separation tasks and are suitable only for single-channel signals.A novel complex-valued T-F mask is proposed that reserves the head-related transfer function(HRTF),customized for binaural sound source localization.In addition,because the convolutional neural network that is exploited to estimate the proposed mask takes binaural spectral information as the input and output,accurate binaural cues can be preserved.Compared with conventional T-F masks that emphasize single speech source–dominated T-F units,HRTFreserved masks eliminate the speech component while keeping the direct propagation path.Thus,the estimated HRTF is capable of extracting more reliable localization features for the final direction of arrival estimation.Hence,binaural sound source localization guided by the proposed T-F mask is robust under noisy and reverberant acoustic environments.The experimental results demonstrate that the new T-F mask is superior to conventional T-F masks and lead to the better performance of sound source localization in adverse environments.

    • 摘要: Sentiment word embedding has been extensively studied and used in sentiment analysis tasks.However,most existing models have failed to differentiate high-frequency and low-frequency words.Accordingly,the sentiment information of low-frequency words is insufficiently captured,thus resulting in inaccurate sentiment word embedding and degradation of overall performance of sentiment analysis.A Bayesian estimation-based sentiment word embedding(BESWE)model,which aims to precisely extract the sentiment information of low-frequency words,has been proposed.In the model,a Bayesian estimator is constructed based on the co-occurrence probabilities and sentiment proba-bilities of words,and a novel loss function is defined for sentiment word embedding learning.The experimental results based on the sentiment lexicons and Movie Review dataset show that BESWE outperforms many state-of-the-art methods,for example,C&W,CBOW,GloVe,SE-HyRank and DLJT1,in sentiment analysis tasks,which demonstrate that Bayesian estimation can effectively capture the sentiment information of low-frequency words and integrate the sentiment information into the word embedding through the loss function.In addition,replacing the embedding of low-frequency words in the state-of-the-art methods with BESWE can significantly improve the performance of those methods in sentiment analysis tasks.

    • 摘要: Circular RNAs(circRNAs)have covalently closed loop structures at both ends,exhibiting characteristics dissimilar to those of linear RNAs.Emerging evidence suggests that aberrantly expressed circRNAs play crucial roles in hepatocellular carcinoma(HCC)by affecting the proliferation,apoptosis and invasive capacity of HCC cells.Certain circRNAs may be used as biomarkers to diagnose and predict the prognosis of HCC.Therefore,circRNAs are expected to become novel biomarkers and therapeutic targets for HCC.Herein,we briefly review the characteristics and biological functions of circRNAs,focusing on their roles in HCC to provide new insights for the early diagnosis and targeted therapy of HCC.

    • 摘要: Objective:Knee osteoarthritis(KOA)is a common chronic joint disease that usually occurs in older people,and injuries articular cartilage and cartilage proliferation. Studies have shown that platelet-rich plasma(PRP),as one of non-surgical treatments,is used to alleviate the symptoms of patients with KOA,becoming popular with doctors. In this study,the purpose was to investigate the effects of platelet-rich plasma on the level of pain and function in patients with KOA. Methods:The Pubmed,Cochrane Library,Embase,and Medline were investigated from 2005 through May 2020. All randomized double-blind controlled trials on the effect of PRP injections for KOA were included. Outcome indicators included the Western Ontario and McMaster Universities Arthritis Index(WOMAC)and the Knee Osteoarthritis Outcome Score(KOOS)for function,the visual analogue scale(VAS)for pain,and adverse events(AEs). Review Manager 5.3 was used to calculate these outcome indicators. Results:In this study,(1)Seven studies met the eligibility criteria,including 250 patients in the experiment(PRP)group and 230 patients in the control(no PRP)group.(2)Meta-analysis results showed that the WOMAC scores were differed significantly between the PRP and no PRP groups at the 6 months follow-up(MD=9.63,95%CI:3.29 to 15.96,P=0.003),and 12 months follow-up(MD=3.85,95%CI:2.66 to 5.04,P0.05).(3)Results showed that PRP has significant advantages in pain relief and functional improvement. Conclusion:In improving knee function and relieving pain,PRP showed superiority to other non-surgical treatments in long-term follow-up from well-designed double-blind trials. Due to the limited quality and data,more high-quality multi-center studies are required.

    • 摘要: For a long time since their discovery by Christian de Duve in the 1950s,lysosomes have been referred to almost exclusively as passive garbage bags;the endpoint in the degradation of intra-and extracellular cargo.The catabolic function of lysosomes is accomplished by an array of more than 60 acid hydrolases,which together break down a wide variety of biological macromolecules,including proteins,lipids,carbohydrates,and nucleic acids,for reutilization in the metabolic processes of the cell.For their optimal function,these enzymes require an acidic intraluminal pH of~4.5,which is maintained by the joint action of a proton pump,the vacuolar H+-ATPase,and several ion channels embedded in the lysosomal limiting membrane.

    • 摘要: BACKGROUND Growing evidence have demonstrated that thyroid hormones have been involved in the processes of cardiovascular metabolism.However,the causal relationship of thyroid function and cardiometabolic health remains partly unknown.METHODS The Mendelian randomization(MR)was used to test genetic,potentially causal relationships between instrumental variables and cardiometabolic traits.Genetic variants of free thyroxine(FT4)and thyrotropin(TSH)levels within the reference range were used as instrumental variables.Data for genetic associations with cardiometabolic diseases were acquired from the genome-wide association studies of the FinnGen,CARDIoGRAM and CARDIoGRAMplusC4D,CHARGE,and MEGASTROKE.This study was conducted using summary statistic data from large,previously described cohorts.Association between thyroid function and essential hypertension(EHTN),secondary hypertension(SHTN),hyperlipidemia(HPL),type 2 diabetes mellitus(T2DM),ischemic heart disease(IHD),myocardial infarction(MI),heart failure(HF),pulmonary heart disease(PHD),stroke,and non-rheumatic valve disease(NRVD)were examined.RESULTS Genetically predicted FT4 levels were associated with SHTN(odds ratio=0.48;95%CI=0.04−0.82,P=0.027),HPL(odds ratio=0.67;95%CI=0.18−0.88,P=0.023),T2DM(odds ratio=0.80;95%CI=0.42−0.86,P=0.005),IHD(odds ratio=0.85;95%CI=0.49−0.98,P=0.039),NRVD(odds ratio=0.75;95%CI=0.27−0.97,P=0.039).Additionally,genetically predicted TSH levels were associated with HF(odds ratio=0.82;95%CI=0.68−0.99,P=0.042),PHD(odds ratio=0.75;95%CI=0.32−0.82,P=0.006),stroke(odds ratio=0.95;95%CI=0.81−0.97,P=0.007).However,genetically predicted thyroid function traits were not associated with EHTN and MI.CONCLUSIONS Our study suggests FT4 and TSH are associated with cardiometabolic diseases,underscoring the importance of the pituitary-thyroid-cardiac axis in cardiometabolic health susceptibility.

    • 摘要: Gastric cancer(GC)is the fourth leading cause of cancer-related death.The occurrence and development of GC is a complex process involving multiple biological mechanisms.Although traditional regulation modulates molecular functions related to the occurrence and development of GC,the comprehensive mechanisms remain unclear.Ultraconserved region(UCR)refers to a genome sequence that is completely conserved in the homologous regions of the human,rat and mouse genomes,with 100%identity,without any insertions or deletions,and often located in fragile sites and tumour-related genes.The transcribed UCR(T-UCR)is transcribed from the UCR and is a new type of long noncoding RNA.Recent studies have found that the expression level of T-UCRs changes during the occurrence and development of GC,revealing a new mechanism underlying GC.Therefore,this article aims to review the relevant research on T-UCRs in GC,as well as the function of T-UCRs and their regulatory role in the occurrence and development of GC,to provide new strategies for GC diagnosis and treatment.
