伯克霍尔德氏菌胞外酯酶催化己酸乙酯合成的研究

         

摘要

从变质的食用油中分离得到1株具有产己酸乙酯酯化酶能力的伯克霍尔德氏菌(Burkholderia sp.)X10,并对其发酵条件和酯化条件进行了优化。结果表明,该菌株最佳发酵条件为:淀粉4%,酵母膏3%,橄榄油4%,发酵液初始pH值6.5,培养温度30℃,转速180 r/min。最佳酯化条件为:己酸添加量2%,酯化温度35℃,酯化液初始pH4.6,振荡方式为连续振荡。优化条件下酯化酶活力由1.79 U/mL提高到3.53 U/mL,较优化前增加了97.21%。%A Burkholderia sp. strain X10 with esterifying enzyme-producing capability was obtained from bad cooking oil. Its fermentation and esterification conditions were optimized. The experimental results revealed its optimum fermentation conditions as follows: 4 % starch,3 %yeast extract,4%olive oil, the initial pH value of the fermenting liquid was 6.5, culture temperature was at 30℃, and the rotation speed was at 160 r/min;its optimal esterification conditions as follows:2%caproic acid,esterifying temperature was at 35℃, the initial pH value of esterify-ing liquid was 4.6, and continuous shock was adopted. Under the optimized conditions, esterifying activity got improved from 3.58 U/mL to 7.06 U/mL, 97.21%higher than that before the optimization.
