豌豆蛋白挤出物抗氧化活性评价及研究

     

摘要

The purpose of this paper was to optimize enzymatic conditions of pea protein by res-ponse surface methodology,to obtain the hydrolysate with the highest radical scavenging ratio and set an acc-urate prediction model.The enzymatic conditions of pea protein by neutral protease were optimized to sca-venging ratio for DPPH and · OH by response surface methodology,after extrusion processing.the best enzymatic conditions list as follows:enzyme amount 11.8%(E/S),temperature 37.5 ℃,pH 6.90,subs-trate concentration 8.00%(g/mL).Under this condition,the scavenging ratio of DPPH and · OH were 95.4% and 88.3 %,respectively.The model prediction is accurate and reliable.The hydrolysis of pea protein by neutral protease could obtain hydrolysate of both high DPPH and high · OH scavenging ra-tio.The model can predict the free radical scavenging activity of the product accurately,also can determine the enzymatic conditions according to the requirements of the product,and provide the basis and reference value for the related production and development of the process product.%本研究旨在应用响应曲面实验设计优化豌豆蛋白酶解条件,以期得到最高的自由基清除酶解产物和准确的酶解预测模型.以豌豆蛋白为原料,经过挤压处理后,选用中性蛋白酶进行酶解,利用响应曲面设计优化豌豆蛋白最佳酶解条件及在此条件下的DPPH及·OH清除率.酶解最佳条件:加酶量11.8% (E/S)、温度37.5℃、pH6.90、底物浓度8.00% (g/mL).在此条件下,得到DPPH清除率、·OH清除率分别为95.4%、88.3%,经验证,所建模型预测准确可靠.豌豆蛋白经中性蛋白酶酶解可获得既具有高DPPH又具有高·OH清除率的酶解产物.所建模型可准确预测产物自由基清除活性,也可以根据产物要求确定酶解条件,为相关生产和工艺产品开发提供依据和参考价值.
