酶联免疫法与LC-MS/MS法检测鸡肉中呋喃它酮代谢物

     

摘要

ELISAandLC-MS/MSfortheanalysisoffuraltadonemetabolite (AMOZ)residuesinchicken meat were studied. The sensibility, accuracy and repeatability of ELISA method were compared to those of LC-MS/MS method. The detection limit of ELISA and LC-MS/MS method could reach to 0.1μg/kg respectively. When the samples were spiked with 0.2, 0.4 and 0.8μg/kg of AMOZ,the average recovery rates of ELISA were 75.0%-94.6%, the relative standard deviation (RSD) was 7.9%-14.3%. And the average recovery rates of LC-MS/MS were 89.5%-92.3%, while the relative standard deviation (RSD) was 8.0%-10. 3%. The results were basically the same to the positive sample.TheresultsshowedthatELISAwasarapidandexactmethodwithgood reproducibility suitable for the screen ofAMOZpositivesamples, LC-MS/MSwassensitiveandsuitablefordeterminationofpositivesamples.%  运用酶联免疫法和LC-MS/MS法检测鸡肉中呋喃它酮代谢物的残留,对两种方法的灵敏度、准确度、重现性等参数进行了比较.结果显示,酶联免疫法和LC-MS/MS法的检测限可以达到0.1μg/kg,对鸡肉样品分别添加0.2、0.4、0.8μg/kg三个浓度水平的呋喃它酮代谢物时,酶联免疫法平均回收率为75.0%~94.6%,变异系数为7.9%~14.3%;LC-MS/MS法的平均回收率为89.5%~92.3%,变异系数为8.0%~10.3%,两种检测方法对阳性样品的检测结果一致.酶联免疫法可以作为初筛方法进行呋喃它酮代谢物的快速筛选,LC-MS/MS法适用于对筛选样品的确证和精确定量.
