液相色谱法测定燕窝中唾液酸含量研究

         

摘要

Objective: To establish a method for determination of content of sialic acid in edible bird's nest by HPLC.Methods:300SCX cation exchange column, 4.6×250mm, particle size 5 μm, mobile phase: acetonitrile -0.1% phosphoric acid solution (90∶10), column temperature:30℃, the flow rate: 1.0ml / min, injection volume:10μl, detection wavelength:205 nm.Results:The retention behavior of the sialic acid in different columns were studied, the SCX cation exchange column has the best retain effect for sialic acid; linear range: 4~100 μg/ml, y=5.08517x+0.0023224, r=0.99996.The average recoveries of bird’s nest is 96.8%, RSD=1.16%;The average recoveries of bird's nest with rock sugar is 96.4%, RSD=1.00%.Conclusions:The method can be used for quality control of bird's nest.and it’s products.%目的:建立液相色谱法测定燕窝中唾液酸含量的方法.方法:300SCX阳离子交换色谱柱,4.6×250mm,粒径5μm,流动相:乙腈-0.1%磷酸水溶液(90:10),柱温:30℃,流速:1.0ml/min,进样量:10μl,检测波长:205nm.结果:系统研究了唾液酸在不同色谱柱中的保留行为,确定SCX型阳离子交换柱对唾液酸有最好的保留;线性范围:4~100μg/ml,线性方程:y=5.08517x+0.0023224,r=0.99996;燕窝平均回收率为96.8%,RSD=1.16%;冰糖燕窝平均回收率为96.4%,RSD=1.00%.结论:本法可用于燕窝及燕窝制品的质量控制.
