目的:采用基于超高效液相色谱与串联四级杆飞行时间质谱仪(UPLC/Q-TOF-MS)联用的代谢组学技术,研究代谢综合征痰瘀互结证动物模型血清中小分子代谢物的变化.方法:模型组大鼠复制成痰瘀互结证代谢综合征,运用UPLC/Q-TOF-MS技术检测空白对照组和模型组血清样本中的小分子代谢产物,对所得到的代谢指纹图谱进行判别分析,找出具有差异的特征代谢物并追溯其来源.结果:鉴定出的潜在生物标志物主要有溶血磷脂酰胆碱、苯丙氨酸、酪氨酸、十四烷酸、硬脂酸、D-葡糖酸、前列腺素B1等内源性代谢物.结论:痰瘀互结证代谢综合征的发生与糖类、脂类、氨基酸代谢紊乱有关,这些体内的小分子化合物可能是痰瘀互结证代谢综合征中医证候的物质基础.%Objective:We analyzed the changes of endogenous metabolites in serum of rats with the syndrome of phlegm and blood stasis syndrome based on UPLC/Q-TOF-MS.Methods:We used UPLC/Q-TOF-MS technology and detected small molecule metabolites in plasma and identified the differences in characteristics of metabolites and found some biomarkers,so as to identify biomarkers and its origins and metabolic pathway.Results:The identified potential biomarkers include LysoPE、Phenylalanine、Tyrosine、14-alkyl acid、Stearic acid、D-glucose、Prostaglandin B1.Conclusion:The occurrence of metabolic syndrome of phlegm and blood stasis syndrome is related to the disorder of carbohydrate,lipid and amino acid metabolism.
