碱性蛋白酶对蛹虫草蛋白质水解的工艺技术优化研究

         

摘要

Using Cordyceps militaris protein powder as raw material,alkaline protease was used for enzymatic treatment and the enzymatic hydrolysis process was optimized.Using hydrolysis degree and enzymolysis yield as evaluation indexes,the hydrolysis process of C.militaris was optimized by orthogonal experiments.When the processing conditions were substrate concentration 8%,alkaline protease addition 1.3%,enzymolysis time 10 h,temperature 53 ℃,the hydrolysis degree was the highest of 15.01%;When the processing conditions were substrate concentration 8%,alkaline protease 1.9%,enzymolysis time 8 h,temperature 56 ℃,the enzyme hydrolysis yield was the highest of 75.27%.After alkaline protease hydrolysis,filtration,drying and other processes,the molecular mass of the hydrolysates was small,hydrolysis residue was little,and the enzyme hydrolysis yield was high,which could be applied in industrialization.%以蛹虫草蛋白粉为原料,利用碱性蛋白酶进行酶解处理,并对其酶解工艺进行优化研究.以水解度和酶解得率为评价指标,采用正交试验结果对蛹虫草水解工艺进行优化.当底物浓度为8%、碱性蛋白酶的加酶量为1.3%、酶解时间10h、酶解温度为53℃时的水解度最高,为15.01%;当底物浓度为8%、碱性蛋白酶的加酶量为1.9%,酶解时间8h、酶解温度为56℃时的酶解得率最高,为75.27%.经碱性蛋白酶酶解、过滤、干燥等工艺处理后,酶解产物的分子质量小,酶解剩余物少,酶解得率高,可以进行产业化推广应用.
