To assess the clastogenic effects of a diglyceride preparation containing conjugated linoleic acid (DG+CLA) an in vivo micronucleus test was performed using ICR mice. 3 doses of DG+CLA (500, 1000 and 1000 mg/kg) were administered orally, Mitomycin C (positive control, 2 mg/kg, administered intraperitoneally) and negative control (olive oil, 10 ml/kg, administered orally). A slide preparation was made at 24 h after the 1st treatment with DG+CLA. Micronucleated polychromatic erythrocyte (MNPCE) of 2000 polychromatic erythrocytes (PCE) were counted. The number of aberrant cells was not increased by any of the 3 doses of DG+CLA orally administered. There were no clinical signs connected with administration of DG+CLA. These results indicate that DG+CLA is not capable of inducing micronuclei in vivo in mice cells and thus has no genotoxicity in the micronucleus.
