阿魏酸诱导胃癌SGC-7901细胞凋亡及对COX-2、Survivin、XIAP和p53的影响

         

摘要

Objective: To discuss the effects of ferulic acid on the proliferation of human gastric cancer SGC-7901 cells and its influence on the expression of apoptosis-related gene. Methods: Different concentrations of ferulic acids were used to intervene human gastric cancer SGC-7901 cells cultured in vitro, after acting on for 24, 48 and 72 hours, MTT method was used to detect cellular proliferation; after different concentrations (0, 5, 7.5 and 10 mg/m L) of ferulic acids acting on SGC-7901 cells for 48 hours, flow cytometry was adopted to detect cellular apoptosis, RT-q PCR and Western-blot were uased to measure m RNA and protein expressions of COX-2, Survivin, XIAP and p53. Results: Compared with the control group, ferulic acids in the concentrations of 7.5, 10 and 12.5 mg/m L could decrease OD values of SGC-7901 cells, ferulic acids in the concentrations of 5, 7.5 and 10 mg/m L after acting on SGC-7901 cells for 48 hours could induce cellular apoptosis, ferulic acids in the concentrations of 5, 7.5 and 10 mg/m L could increase m RNA and protein expression of p53, decrease m RNA and protein expressions of COX-2, Survivin and XIAP. Conclusion: Ferulic acid could effectively inhibit the proliferation of SGC-7901 cells, induce its apoptosis, it could increase m RNA and protein expression of gastric cancer p53, decrease m RNA and protein expression of COX-2, Survivin and XIAP, and produce anti-tumor effects.%目的:探讨阿魏酸对人胃癌SGC-7901细胞增殖的影响及其对凋亡相关基因表达的影响.方法:不同浓度(0、1.25、2.5、5、7.5、10、12.5 mg/mL)阿魏酸作用体外培养人胃癌SGC-7901细胞24、48、72小时后,MTT法检测细胞增殖;不同浓度(0、5、7.5、10 mg/mL)阿魏酸作用SGC-7901细胞48小时,流式细胞仪检测细胞凋亡,RT-q PCR和免疫印迹法(Western-blot)检测环氧化酶2(COX-2)、生存素(Survivin)、X连锁凋亡抑制蛋白(XIAP)和p53的mRNA和蛋白表达.结果:与对照组比较,7.5、10、12.5 mg/mL阿魏酸使SGC-7901细胞的OD值降低,5、7.5、10 mg/mL阿魏酸作用SGC-7901细胞48小时后均能诱导其凋亡,5、7.5、10 mg/mL阿魏酸均能上调p53的mRNA和蛋白表达,下调COX-2、Survivin和XIAP的mRNA和蛋白表达.结论:阿魏酸能有效抑制SGC-7901细胞增殖,并能诱导其凋亡,阿魏酸能上调胃癌细胞p53的mRNA和蛋白表达,下调COX-2、Survivin和XIAP的mRNA和蛋白表达,发挥抗肿瘤作用.
