红花水煎液对系统性硬皮病小鼠皮肤及肺组织纤维化的影响及机制

         

摘要

目的 探讨红花水煎液对系统性硬皮病(SSc)小鼠皮肤及肺组织纤维化的影响及其作用机制.方法 32只BALB/C小鼠随机分为空白组、模型组、强的松组、红花组,每组8只.空白组背部注射生理盐水,其余三组均背部注射博来霉素制备SSc模型;造模同时空白组和模型组给予生理盐水0.2 mL/d灌胃,强的松组给予强的松溶液0.2 mL/d(含强的松0.09 mg)灌胃,红花组给予红花水煎液0.2 mL/d(含红花30 mg)灌胃,均连续给予28天.取各组背部注射区皮肤和肺组织行HE和Masson染色,检测真皮厚度;采用样本水解法检测皮肤和肺组织羟脯氨酸(HYP)含量;采用免疫组化法检测皮肤和肺组织IL-17、IL-6表达;ELISA法检测血清IL-17、IL-6水平.结果 空白组真皮厚度为(295.90±53.44)μm,模型组为(689.01±157.20)μm,强的松组为(562.58±113.74)μm,红花组为(433.80±83.62)μm,组间比较P均<0.05.与空白组比较,其余三组皮肤和肺组织HYP含量均升高;与模型组比较,强的松组、红花组皮肤和肺组织HYP含量均降低,红花组降低更显著;组间比较P均<0.05.与空白组比较,其余三组皮肤和肺组织IL-17、IL-6表达及血清IL-17、IL-6水平均升高;与模型组比较,强的松组、红花组皮肤和肺组织IL-17、IL-6表达及血清IL-17、IL-6水平均降低,红花组降低更显著;组间比较P均<0.05.结论 红花水煎液可改善SSc小鼠皮肤及肺组织纤维化;抑制IL-17、IL-6表达,减轻免疫炎症反应可能是其作用机制之一.%Objective To study the effects of safflower decoction on the skin and lung fibrosis of mice with systemic scleroderma ( SSc) and its mechanism.Methods Thirty-two BALB/C mice were randomly divided into the blank group, model group, prednisone group and safflower group, 8 mice in each group.The blank group was injected with normal sa-line, and the other three groups were treated with bleomycin ( BLM) at the back to establish the SSc models.At the same time, the blank group and model group were given normal saline (0.2 mL/d), the prednisone group was given prednisone (0.2 mL/d), safflower group was given safflower decoction (0.2 mL/d), and all groups were treated for 28 days.After 28 days, all mice were decapitated and the blood, back skin of the BLM injection part and lungs were collected.The patho-logical changes were observed by HE and Masson staining.After that, all the tissue slices were taken to measure the dermal thickness, and we detected the hydroxyproline ( HYP) content in the skin and lung tissues by sample hydrolysis method. The expression of IL-17 and IL-6 in the skin and lung tissues was detected by immunohistochemisty.The levels of IL-17 and IL-6 in serum was determined by ELISA.Results The dermal thickness was (295.90 ±53.44) μm in the blank group, (689.01 ±157.20) μm in the model group, (562.58 ±113.74) μm in the prednisone group, and (433.80 ± 83.62) μm in the safflower group (all P<0.05).Compared with the blank group, the contents of HYP in the skin and lung tissues of the other three groups were increased;compared with the model group, the contents of HYP in the skin and lung tissues were decreased in the prednisone group and safflower group, and the decrease in the safflower group was more significant, and significant difference was found between groups (all P<0.05).Compared with the blank group, the ex-pression of IL-17 and IL-6 in the skin and lung tissues and the levels of IL-17 and IL-6 in the serum of the other three groups were increased;compared with the model group, the expression of IL-17 and IL-6 in the skin and lung tissues and the levels of IL-17 and IL-6 in the serum were decreased in the prednisone group and safflower group, and the decrease in the safflower group was more significant ( all P<0.05) .Conclusions Safflower decoction can improve lung and skin fi-brosis in SSc mice.Inhibiting the expression of IL-17 and IL-6, and alleviating immune inflammatory reaction may be one of its mechanisms.
