大豆异黄酮对脑缺血再灌注大鼠海马神经再生的影响

         

摘要

目的 观察大豆异黄酮对脑缺血再灌注大鼠海马神经再生的影响.方法 线栓法建立大鼠脑缺血再灌注模型,分别给予大豆异黄酮(SI)、7-硝基吲哚(7-NI)或联合给药(SI+7-NI),BrdU免疫荧光法标记新生海马神经细胞,免疫组化法检测海马区ONOO-的反应产物硝基酪氨酸(NT).结果 SI组、7-NI组和SI+7-NI组大鼠海马区BrdU+细胞数增加更显著,NT的数量明显减少(P<0.05,P<0.01).结论 长期给予大豆异黄酮可以使脑缺血再灌注时病理性的ONOO-生成减少,并促进脑缺血组织的海马神经再生,这对脑缺血损伤有潜在的应用价值.%OBJECTIVE To observe the effect of soy isoflavones on the hippocampus neurogenesis of rats with cerebral ischemia and reperfusion.METHODS Models of rats with cerebral ischemia and reperfusion were built by the method of middle cerebral artery occlusion, which were treated with soy isoflavones(SI), 7-Nitroindole(7-NI) or coadministration of SI and 7-NI(SI+7-NI), respectively.Then neonatal hippocampus nerve cells were labeled with BrdU by immunofluorescence, and the nitrotyrosine(NT) :and the production of ONOO were detected by immunohistochemisty.RESULTS The amounts of BrdU+ nerve cells and the quantity of NT in the zone of hippocampus in the model group were increased significantly compared with control group(P<0.01 ).The amounts of BrdU+ nerve cells in the zone of hippocampus in the SI, 7-NI, or SI + 7-NI group were increased significantly compared with model group( P<0.01 ), while the quantity of NT in the zone of hippocampus in the SI, 7-NI, or SI +7-NI group were decreased significantly compared with model group(P<.0.05, P<0.01).CONCLUSION The pathologic production of ONOO cart be decreased and the hippocampus neurogenesis can be promoted with long-term low-dose administration of soy isoflavones for rats with cerebral ischemia and reperfusion.It shows that soy isoflavones have the potential applying values on the therapy of cerebral ischemic damages.
