海洋真菌MF-08产壳聚糖酶诱导条件及酶学性质分析

         

摘要

The influence of different shaker's rotation speeds,carbon source, carbon source concentration and induced carbon source for the marine fungal MF-08 producing chitosanase were researched to increase enzyme activity,and the characteristics of chitosanase was researched. The results showed that the optimal fermentation conditions was 27℃,150 r/min,1.5% concen-tration,0.01% glucosamine, fermenting in seawater medium 72 h. The optimum temperature and pH for Chitosanase were 40℃and 5.2,respectively. And the chitosanase was stable at 0~40℃ and 4.0~6.0, respectively. The enzyme activity was inhibited by Cu2+,Fe3+ and Hg2+. Under the optimal cultivation conditions,the enzyme activity could reach 1.769 U/mL. After optimiza-tion,the enzyme activity could be increased by 7.56.%通过研究不同摇床转速、不同碳源和碳源浓度、不同诱导碳源对海洋真菌MF-08产壳聚糖酶的影响,提高其产酶活性,并对海洋真菌MF-08产壳聚糖酶进行部分酶学性质研究.结果表明,最佳发酵条件为27℃,150 r/min,1.5%蔗糖,0.01%氨基葡萄糖,海水培养基发酵72 h.该壳聚糖酶的最适作用温度为40℃,最适pH 5.2,酶活在pH 4.0~6.0范围内和0~40℃相对稳定;Cu2+、Fe3+和Hg2+对该酶活具有明显的抑制作用.经过培养条件的优化,该菌的最高产酶活性达到1.769 U/mL,与优化前相比提高了7.56倍.
