表达 US3腺病毒载体对混合淋巴细胞和 NK 细胞反应中γ干扰素和穿孔素表达的影响

         

摘要

目的:探讨表达US3基因的重组腺病毒载体介导其转染供体外周血单个核细胞(PBMCs)的免疫逃逸活性。方法首先构建表达US3基因的腺病毒载体,扩增纯化后转染供体PBMCs,利用混合淋巴细胞和NK细胞反应,通过检测反应中γ干扰素和穿孔素的表达量,观察表达US3基因重组腺病毒载体的免疫活性。结果表达US3基因重组腺病毒载体r-US3能够明显降低混合淋巴细胞和NK细胞反应中细胞因子γ干扰素和穿孔素的产量,抑制混合淋巴细胞和NK细胞反应活性。结论表达US3基因腺病毒载体在一定程度上能够介导其转染供体PBMCs的免疫逃逸,降低机体免疫系统对其转染细胞的排斥反应。%Objective To investigate the activity of immune evasion induced by a recombinant adenovirus vector expressing the US3 gene in transfected peripheral blood mononuclear cells (PBMCs).Methods An adenovirus vector ex-pressing the US3 gene was established , multiplied and purified before being transfected into peripheral blood mononuclear cells ( PBMCs) .Immunological activities of US 3+cells were determined by testing the expression levels of interferon -γ( INF-γ) and perforin in mixed lymphocyte and NK cells reaction .Results r-US3 could significantly attenuate mixed lymphocyte and NK reaction by decreasing production of INF -γand perforin.Conclusion The adenovirus vector ex-pressing US3 gene may induce immune evasion in PBMCs with the transfected gene , attenuating rejection of the host im-mune system against transfected cells .
