高效液相色谱法测定饮料、泡菜中的赤藓红

             

摘要

To establish a rapid method for determination of erythrosine in drinks and pickles by high-performance liquid chromatography (HPLC). The chromatography condition were C18 column as the analytica column, detection wavelength at 530nm, column temperature at 35℃, 0.005mol/L ammonium acetate solution and methanol as mobile phase, flow rate of 1.0mL/min and injection volume of 10μL. The test sample after ultrasonic extraction, precipitat-ed protein, the high speed centrifugation, filtration and finally directly by HPLC. The concentration of erythrosine showed linearity between 4-40μg/mL, the correlation coefficient was 0.9999. The average recovery was from 89.0%to 98.8%, and the relative standard deviation was from 0.78% to 1.32%. The method is simple, rapid, accurate, high-sensitivity, less interference, it is suitable for determination of erythrosine in drinks and pickles.%建立一种快速处理样品并检测样品中赤藓红含量的高效液相色谱法。选择C18色谱柱为分析柱;检测波长:530nm;柱温:35℃;流动相:0.005mol/L乙酸铵溶液-甲醇(42+58);流速为1.0mL/min;进样量为10μL。待测样品加水经超声波提取,沉淀蛋白质,高速离心后经滤膜过滤后直接上机测定。赤藓红浓度在4-40μg/mL之间呈线性关系,相关系数0.9999。平均回收率为89.0-98.8%,相对标准偏差为0.78-1.32%。该法操作简单、快速、准确,灵敏度高、干扰少、适用于饮料和泡菜中赤藓红的测定。
