正交试验优选刺玫果总黄酮提取工艺研究

     

摘要

目的:确定刺玫果总黄酮的最佳提取工艺,为刺玫果总黄酮分离纯化提供数据支持。方法以总黄酮含量为主要指标、总黄酮转移率为次要指标,采用正交试验设计优选提取参数,以金丝桃苷为对照,采用紫外分光光度法测定刺玫果提取物中总黄酮的含量。结果优选工艺为刺玫果药材加6倍量60%乙醇回流提取,提取2次,每次1.5 h。结论优化选择的提取工艺较稳定,重现性好,可为生产实践提供理论依据,并为刺玫果总黄酮分离纯化提供数据支持。%Objective To determine the optimum extraction process of total flavonoids in Rosa fruit.Methods With the content of total flavonoids as the main indicator and the transport rate of total flavonoids as the secondary indicator, the extraction parameters were optimized by orthogonal design. With hyperoside as control, the content of total flavonoids in Rosa fruit was determined by UV spectrophotometry.Results The optimal extraction process was as follows: the refluxing extraction was performed with 60 % ethanol, with the ratio of material to liquid of 1:8. The extraction was repeated for two times, 1 h for each time.Conclusion The optimal extraction process can be stable and reproducible, and can provide theoretical basis for production practice, as well as data support for the separation and purification of total flavonoids in Rosa fruit.
