氯化三乙基锡抑制C6胶质瘤细胞增殖及机制

         

摘要

Objective To study the inhibition mechanism of triethyltin chloride (TETC) on proliferation of C6 glioma cells in vitro. Methods Inhibitory rate of TETC on the proliferation of C6 glioma cells , apoptotic rate of C6 glioma cells and expression of Bcl-2 and Bax protein were tested by MTT assay, fluorescent TUNEL assay and Western blotting analysis at 48 h after treated by different concentrations of TETC on C6 glioma cells, respectively. Results The proliferation of C6 glioma cells was inhibited by TETC in a dose-dependent manner. TETC induced apoptosis of C6 glioma cells and the apoptotic rate was increased in a duse-dependent manner. The expressions of Bcl-2 and Bax protein were down-regulated and up-regulated, respectively. Conclusions TETC could inhibit the proliferation of C6 glioma cells in vitro. TETC could induce the apoptosis of C6 glioma cells, decrease the expression of Bcl-2 protein, increase the expression of Bax protein, lead to apoptosis of C6 glioma cells, and these maybe the mechanism of anti-proliferation of TETC.%目的 探讨氯化三乙基锡(TETC)对C6胶质瘤细胞体外增殖抑制作用的机制.方法 采用MTT法、荧光原位末端标记方法和Western印迹方法,分别检测不用浓度TETC作用C6胶质瘤细胞48 h后细胞增殖抑制率、细胞凋亡率和Bcl-2和Bax蛋白的表达水平.结果 TETC对C6胶质瘤细胞具有抑制增殖作用,增殖抑制率呈剂量依赖性;TETC可诱导C6胶质瘤细胞凋亡,细胞凋亡率呈剂量依赖性上升;TETC可下调Bcl-2和上调Bax蛋白表达水平.结论 TETC可抑制C6胶质瘤细胞体外增殖并诱导其凋亡,机制可能与其下调Bcl-2和上调Bax蛋白表达有关.
