葡萄籽原花青素的纤维素酶辅助提取工艺优化

         

摘要

本文首先在单因素试验的基础上,采用Box - Behnken设计对葡萄籽原花青素的纤维素酶辅助提取工艺中的pH、酶浓度和酶解温度3因子的最优化组合进行了定量研究,建立并分析了各因子与得率关系的数学模型;然后与传统提取方法进行了比较.结果表明:最佳的工艺条件为pH 4.4、酶浓度0.4%和酶解温度47.8℃.经试验验证,在此条件下,得率为3.37%,与理论计算值3.35%基本一致,说明回归模型能较好地预测葡萄籽中原花青素的提取得率;与传统提取方法相比,纤维素酶辅助提取时间短,得率高.%The influences of pH, enzyme concentration and enzymolysis temperature on the yield of the proanthocyani-dins from grape seeds by cellulase - assisted extraction technique were studied. Box - Behnken experimental design was applied based on the single factor experiment. The mathematical model was established in order to analyze the relationships between the studied factors and the proanthocyanidin extraction yield. Then the cellulase - assisted extraction and traditional extraction techniques were compared. Based on the canonical analysis, the optimum extraction conditions were: pH 4. 4, enzyme concentration 0. 4% and enzymolysis temperature 47. 8℃. Under the optimized extraction conditions, the extraction yield was 3. 37% , which was very close to the theoretical estimated value of 3. 35%. This shows the regression model can be used to well predict the experiment results. Compared with traditional extraction techniques , cellulase - assisted extraction had higher yield and shorter extraction time.
