Optimization of Ultrasonic Extraction for Citrus Polysaccharides by Response Surface Methodology

The ultrasonic-assisted extraction of polysaccharides from citrus peel was modeled by response surface methodology which adopts a Box-Behnken design with the three-factor-three-level. Based on the single-factor-test, the Box-Behnken design was employed to optimize the extraction variables (extraction temperature, ultrasonic power, and ratio of distilled water to raw material) for the extraction yield of polysaccharides. The statistical analyses showed that the quadratic terms (X12, X22 and X32) and the interaction of X2 with X3 had significant effect on the yield (p < 0.05). The optimized conditions were found as follows: extraction temperature of 50 °C, ultrasonic power of 370 W, ratio of distilled water to raw material of 31:1 mL/g. Under these conditions, the extraction yield of polysaccharides was 30.77 ± 0.48% (n = 3), which was closed to the model predicted value of 30.72%. This study can provide the theoretical guidance for the industrial extraction of polysaccharides and improve the utilization of citrus peels.
