    • 摘要: Background:Nicotine administration can generate severe oxidative stress and lipid peroxidation.Royal jelly,with its antioxidant properties,acts as a scavenger of reactive oxygen species.This study describes the apitherapy effects of royal jelly on testicular damage following nicotine administration.Methods:Forty-eight male BALB/c mice were divided into 8 groups(n=6):saline,3 different doses of royal jelly(100,150,and 200 mg/kg body weight(BW)per day),nicotine(1.5 mg/kg),and 3 different groups of Nic+Roy(1.5 mg/kg of Nic+100,150,and 200 mg/kg BW per day of royal jelly).Nicotine was administrated intraperitoneally,and royal jelly was prescribed orally for 10 consecutive days.Serum levels of hormones(testosterone,luteinizing hormone,and follicle-stimulating hormone),total antioxidant capacity,nitric oxide(NO)status,malondialdehyde levels,sperm DNA fragmentation,sperm parameters,histopathological changes(H&E staining),immunohistochemistry against apoptotic proteins,and gene expression of Bcl-2,p53,Caspase-3,and Nrf2(real-time PCR)were assessed to evaluate the molecular and histological changes.Results:Hormone levels,sperm parameters,and status of antioxidants were decreased significantly(p<.05)following nicotine administration.Moreover,royal jelly treatment normalized hormonal and antioxidant characteristics,decreased apoptotic gene expression,increased Nfr2 gene expression,and restored histopathological alteration to the physiological status significantly(p<.05).Conclusion:Royal jelly upregulates the antioxidant status,inhibits the mitochondrialdependent apoptosis pathway,and increases the rate of proliferation.This therapeutic agent effectively protected the testis against nicotine-associated damages by antioxidant and anti-apoptotic effects.

    • 摘要: An increase in oxidative stress plays a key role in neurotoxicity induction and cell death, which leads to neurodegenerative diseases such as Parkinson’s disease and Alzheimer’s disease. Cyanidin-3-glucoside (C3G) is a common anthocyanin and shows antioxidant activity in neuronal cells. Silent information regulator 2-related protein 1 (Sirt1) regulates antioxidant and anti-inflammatory effects. However, the effects of C3G on Sirt1 in neuronal cells remain unclear. This study evaluated the effect of C3G on Sirt1 expression and activity in human neuroblastoma (SH-SY5Y) cells. In the study, C3G increased the expression of Sirt1 and Sirt1 activity in SH-SY5Y cells. Additionally, C3G increased the expression of nuclear factor erythroid 2-related factor 2, a vital transcription factor for regulating the expression of antioxidant genes, as well as antioxidant enzymes such as superoxide dismutase and catalase. Moreover, C3G protected SH-SY5Y cells from oxidative stress. These results suggest that C3G decreased oxidative stress-induced cell injury by increasing the expression of Sirt1 and other antioxidant factors. Therefore, C3G might merit further investigation for use in attenuating the progress of neurodegenerative diseases.

    • 摘要: The objective of this work is to dry the mushrooms to evaluate the impact of drying on them. The drying of mushrooms in the oven, in the solar dryer and in the microwave indicated that for Termitomyces sp. the total polyphenol contents were 15.20 mgEAG/gMs for the microwave drying (280 W), 13.61 mgEAG/gMs for the oven drying (60°C), and 9.81 mgEAG/gMs for the solar dryer drying (40°C). For Pleurotus sp., the contents obtained were 13.79 mgEAG/gMs for microwave drying (280 W), 8.36 mgEAG/gMs for oven drying (60°C) and 8.98 mgEAG/gMs for solar drying (40°C). Regarding flavonoids, for Termitomyces sp., this revealed 0.5797 mgECa/gMs for microwave drying (280 W), 0.729 mgECa/gMs for oven drying (60°C) and 0.5671 mgECa/gMs for solar drying (40°C). The flavonoid contents for Pleurotus sp. gave 0.842 mgECa/gMs for microwave drying (280 W), 1.06 mgECa/gMs for oven drying (60°C) and, 0.425 mgECa/gMs for solar drying (40°C). For Termitomyces sp., the levels of free radical scavenging activity varied from 13.02 mg/mL for microwave drying, 25.08 mg/mL for oven drying, and 22.25 mg/mL for solar drying;while for Pleurotus sp., 15.96 mg/mL for microwave drying, 39.90 mg/mL for oven drying, and 31.60 mg/mL for solar drying.

    • 摘要: The antioxidant property of Moringa oleifera leaves and Pleurotus ostreatus on swimming performance and related biochemical parameters were investigated in thirty adult male and female wistar rats. Purpose: The aim of the present study was to evaluate the antioxidant property of M. oleifera and P. ostreatus mixture in specific proportions in wistar rats. Material and Methods: The mushroom species and M. oleifera leaves were cultivated at the biotechnology laboratory of the University Cheikh Anta Diop de Dakar, Senegal. Oxidative stress in rats was induced by swimming. The dietary supplement was composed of a mixture of powders of Moringa oleifera and Pleurotus ostreatus in 2:1 ratio. Three groups received different doses of dietary supplements 500 mg/kg, 1000 mg/kg, and 1500 mg/kg, with one group being the stressed and the other the control group. Parameters in serum such as serum glutamyl oxaloacetate transaminase, serum glutamyl pyruvate transaminase, albumin, Testosterone, creatinine, and, oxidative stress parameters (catalase, reduced glutathione, and malondialdehyde) were measured. Results: Results showed that the dietary supplement has an effect on oxidative stress because it increases the activity of catalase and the level of cellular glutathione in rats. The 500 mg/kg dose would be the most appropriate dose for stressful conditions. The 1000 mg/kg dose would be the most appropriate dose for liver damage. Conclusion: This study shows that the antioxidant properties of M. oleifera leaves and Pleurotus ostreatus are demonstrated by their ability to improve body energy stores and tissue antioxidant capacity. The dietary supplement of M. oleifera leaves and P. ostreatus powders mixture could be good in stressful conditions.

    • 摘要: Background:Enteromorpha prolifera(E.prolifera)polysaccharide has become a promising feed additive with a variety of physiological activities,such as anti-oxidant,anti-cancer,anti-diabetic,immunomodulatory,hypolipidemic,and cation chelating ability.However,whether Enteromorpha polysaccharide-trace element complex supplementation regulates amino acid and fatty acid metabolism in chicken is largely unknown.This study was conducted to investigate the effects of E.prolifera polysaccharide(EP)-Zn supplementation on growth performance,amino acid,and fatty acid metabolism in chicken.Methods:A total of 184 one-day-old Ross-308 broiler chickens were randomly divided into two treatment groups with 8 replicates,12 chickens per replicate,and fed either the basal diet(control group)or basal diet plus E.prolifera polysaccharide-Zinc(400 mg EP-Zn/kg diet).Results:Dietary EP-Zn supplementation significantly increased(P<0.05)the body weight,average daily gain,muscle antioxidant activity,serum HDL level,and reduced serum TG and LDL concentration.In addition,dietary EPZn supplementation could modulate ileal amino acid digestibility and upregulate the mRNA expression of amino acid transporter genes in the jejunum,ileum,breast muscle,and liver tissues(P<0.05).Compared with the control group,breast meat from chickens fed EP-Zn had higher(P<0.05)Pro and Asp content,and lower(P<0.05)Val,Phe,Gly,and Cys free amino acid content.Furthermore,EP-Zn supplementation upregulated(P<0.05)the mRNA expressions of mTOR and anti-oxidant related genes,while down-regulated protein degradation related genes in the breast muscle.Breast meat from EP-Zn supplemented group had significantly lower(P<0.05)proportions ofΣn-3 PUFA,and a higher percentage ofΣn-6 PUFA and the ratio of n-6/n-3 PUFA.Besides,EP-Zn supplementation regulated lipid metabolism by inhibiting the gene expression of key enzymes involved in the fatty acid synthesis and activating genes that participated in fatty acid oxidation in the liver tissue.Conclusions:It is concluded that EP-Zn complex supplementation regulates apparent ileal amino acid digestibility,enhances amino acid metabolism,and decreases oxidative stress-associated protein breakdown,thereby improving the growth performance.Furthermore,it promotes fatty acid oxidation and restrains fat synthesis through modulating lipid metabolism-related gene expression.

    • 摘要: Nuclear factor erythroid-derived 2-like 2(Nrf2)is the master regulator of antioxidant defenses.High-intensity interval training(HIIT)has been proposed as a time-efficient training program and has become a substantial component of modern training program In the present study,we evaluated the effects of sulforaphane(SFN),a dietary isothiocyanate derived from cruciferous vegetables and a potent Nrf2 activator,on Nrf2-mediated antioxidant defense responses of skeletal muscle induced by exhaustive exercise in HIIT mice.Male C57 BL/6 J mice were randomly allocated into control group,HIIT group,and HIIT pretreated with SFN(HIIT+SFN)group.On the third day after completion of a 6-weeks HIIT protocol,an exhaustive treadmill test was conducted in all mice.Mice were intraperitoneally injected with SFN(HIIT+SFN group)or PBS(HIIT and control mice)4 times in 3 days prior to the exhaustive treadmill test.The results indicated that the 6-weeks HIIT protocol did not increase the antioxidative capacity of skeletal muscle during exhaustive exercise.Importantly,SFN treatment improved anti oxidative capacity of skeletal muscle in response to the acute exhaustive exercise by increasing mRNA and nucleoprotein expression of Nrf2 and these genes involved in antioxidant generation and decreasing blood creatine kinase(CK)and 4-hydroxy-2-nonenal(4-HNE)-modified protein levels in the HIIT mice.

    • 摘要: The antioxidant effects of quercetin were studied in vitro and in vivo.In vitro,vitamin C was used as a positive control to evaluate the antioxidant capacity of quercetin in three aspects:scavenging free radicals,protecting biological macromolecules and the total reducing power.In vivo,a total of 240 AA broilers(1-day age)with similar body weight were randomly divided into four groups with six replicates in each group,and 10 broilers in each replicate.The four groups were fed with corn-soybean basal diet supplemented with 0.00%,0.02%,0.04%and 0.06%quercetin to study its effects on antioxidant indexes of AA broilers,and to explore the optimal dose of quercetin as a dietary additive.The results showed that quercetin scavenged superoxide anion,hydroxyl radical and 1,1-diphenyl-2-picrylhydrazyl(DPPH)in vitro,the scavenging effects of quercetin on O_(2)-and•OH first increased and then decreased with the increase of the concentrations(P•OH>O_(2)-.The inhibition of vitelline lipoprotein peroxidation by quercetin was increased with increasing concentrations(P0.05);however,superoxide dismutase(SOD)activity was significantly increased,whereas glutathione peroxidase(GSH-Px)and catalase(CAT)activities were significantly decreased in livers with increasing quercetin(P<0.05).These results suggested that quercetin exhibited strong antioxidant effects in vitro and in vivo.

    • 摘要: This study aimed to investigate the antioxidant effect of soybean milk fermented by a new type of Lactobacillus fermentum(LF-HFY02)by using D-galactose induced aging mice model.Firstly,the optimal fermentation conditions was screened out by detecting the effects of different fermentation temperature and time on the active components and antioxidant activity of soybean milk in viro.And then unfermented soybean milk and the soybean milk fermented by different Lactobacillus was given by gavage to D-galactose-induced aging mouse.The activities of GSH,GSH-Px,SOD,CAT and T-AOC in serum,brain and liver of soybean milk fermented by LF-HFY02 were significantly increased,while the content of MDA and the level of AGEs in hippocampal were significantly decreased compared with D-galactose induced group.Further more,the mRNA expression of GSH and SOD in mouse liver were obviously up-regulated by soybean milk fermented by LF-HFY02.The skin tissue structure of mice in the LF-HFY02 fermented soybean milk group was more complete,the collagen fibers were increased and arranged orderly and liver inflammation has improved compared with the model group.And Western blot analysis showed that LF-HFY02 effectively upregulated EGFR,SOD and GSH protein expression in mouse liver.These findings suggest that LF-HFY02 can effectively prevent D-galactose-induced oxidation and aging in mice,and the effect was even better than that of the Lactobacillus delbruechii subsp.bulgaricus and vitamin C.Thus,LF-HFY02 may be potentially employed as a probiotic strain.In conclusion,soybean milk fermented by LF-HFY02 can increase the content of antioxidant factors and the activity of antioxidant enzymes by regulating gene and protein expression,and finally inhibit the process of tissue cell peroxidation,and improve the oxidative damage of mouse skin and liver.The results could provide a basis for the research and development and industrial production of probiotic-related fermented soybean milk products.

    • 摘要: Objective:To evaluate the antioxidant potential and pancreatic lipase inhibitory action of optimized hydroethanolic extracts of Solanum nigrum.Methods:Optimized extraction for maximum recovery of metabolites was performed using a combination of freeze-drying and ultrasonication followed by determination of antioxidant and antiobesity properties.The ultra-high performance liquid chromatography equipped with mass spectrometry was used to analyze metabolite profiling of Solanum nigrum.Computational studies were performed using molecular docking and electrostatic potential analysis for individual compounds.The hypolipidemic potential of the most potent extract was assessed in the obese mice fed on fat rich diet.Results:The 80%hydroethanolic extract exhibited the highest extract yield,total phenolic contents,total flavonoid contents along with the strongest 2,2-diphenyl-1-picrylhydrazyl scavenging activity,total antioxidant power,and pancreatic lipase inhibitory properties.The 80%hydroethanolic extract not only regulated the lipid profile of obese mice but also restricted the weight gain in the liver,kidney,and heart.The 80%hydroethanolic extract also reduced alanine transaminase and aspartate transaminase concentrations in serum.The effects of plant extract at 300 mg/kg body weight were quite comparable with the standard drug orlistat.Conclusions:Solanum nigrum is proved as an excellent and potent source of secondary metabolites that might be responsible for obesity mitigation.

    • 摘要: Side reaction pathways are observed to occur between antioxidants and peroxide during the processing of multiple resin grades mediated by controlled peroxide-induced degradation of impact polypropylene.In the present work,the reaction mechanism and main by-products between antioxidants and peroxides were investigated.The results demonstrate that peroxides greatly accelerate the decomposition reactions,and the free radicals formed from peroxide decomposition react with,for example,the antioxidant AO1010 to produce dehydrogenation of phenyl propionic unit(s)at the a position(cinnamic acid ester moiety),which generates a conjugated system leading to the increased color of the product.It is the first time to confirm the cinnamic acid moiety's existence and report its NMR data.Further,this work confirms the dehydrogenation mechanism by comparison with different sterically hindered phenolic antioxidants.It also systematically summarizes the oxidation and degradation mechanism of AO168 and AO1010 under air and peroxide environments,respectively.Based on present study,clear guidelines are obtained to improve the quality of polymeric products,especially the appearance and stability,during product development.
