Monoclonal antibodies (MAb) against TNF-BPI (tumour necrosis factor-binding protein I), designated tbp-1 and tbp-2 and secreted by the hybridomas TBP-1 and -2, and their active fragments are new. Also new are (1) hybridomas TBP-1 and-2 (ECACC 91060555 and 91060556, respectively) and (2) sandwich immunoassay kits for determn. of TNF-BPI in a sample. More specifically, TNF-BPI is detected by reaction with at least one MAb to form a binary or ternary complex. In partic. a ternary complex is formed esp. where the 1st. antibody is tbp-1 and the 2nd. is tbp-2. USE/ADVANTAGE - MAb provide an efficient and sensitive method for detecting TNF-BPI in body fluids (serum, plasma or urine) and cell culture supernatants. The method is used to diagnose (and monitor) diseases associated with activation of the TNF-receptor system, specifically septic shock but also bacterial infections, graft vs. host disease, cerebral malaria and cachexia. TNF-BPI can be determined at 200 ng/l with reproducibility better than 10% and the presence of TNF in the sample does not affect the assay, even at pathologically elevated levels.
