RESEARCH REPORT on NSC-135758: PRECLINICAL TOXICITY OF INTRAVENOUS INJECTIONS IN DOGS AND MONKEYS

2,5-Piperazinedione,3,6-bis-(5-chloro-2-piperidyl)dihydrochloride, NSC-135758, was submitted to a preclinical toxicological evaluation in 22 dogs and 10 monkeys (1 male and 1 female per dose level). Four treatment schedules were used with the dogs: single injection of 3.0, 1.5, 0.75, 0.375, or 0.1875 mg/kg; 5 daily injections of 0.375, 0.1875, 0.0937, or 0.0468 mg/kg/day; 5 daily injections followed by 9 days rest repeated for 3 treatment periods at 0.0937 mg/kg/day; and weekly injections for 6 weeks at 0.375 mg/kg/week. The treatment schedule for monkeys was 5 daily injections of 1.5, 0.75, 0.375, 0.1875, or 0.0937 mg/kg/day.
