Vincristine (NSC 67574): Preclinical Toxicologic Evaluation in Monkeys and Dogs Using Weekly Intravenous Injections for Six Weeks.

Vincristine was injected I.V. weekly,for 6weeks into dogs and monkeys. Two of four dogs treated with 0.08mg/kg/week died,one after the fifth treatment and one after the sixth treatment while all dogs treated with 0.04and 0.02mg/kg/weekly survived. One of two monkeys treated with 0.64mg/kg/week was terminated in a moribund condition after the sixth treatment. Monkeys treated with 0.32, 0.16,or 0.08mg/kg/week all survived in good condition. Necrosis of the intestinal nucosa was the limiting toxicity in dogs while the monkeys exhibited a severe peripheral neuropathy at the high dose level. (Author)
