Transplacental Transfer of Indomethacin and Its Pharmacokinetics in Pregnant Rhesus Monkeys

In order to monitor circulating levels of indomethacin, a sensitive method for the determination of this drug in small samples of plasma has been developed. Following the extraction of the indomethacin from plasma and the formation of a pentafluorobenzyl derivative, the method employs the use of electron capture gas-liquid chromatography. An indomethacin level of use 1 nanogram per milliliter in the lowest level detectable by this method in a plasma sample volume of between 25 to 50 microliters. Plasma levels of indomethacin in the maternal system and the fetal system were monitored in time course studies. These studies resulted in a better understanding of the transplacental transfer of indomethacin and its pharmacokinetics in the pregnant rhesus monkey.
