Oviductal epithelial cell culture was used to investigate the effect of ovarian steroids (progesterone and oestradiol-17beta), oxytocin and LH on the local production of PGE2, PGF2alpha and endothelin-1 in cow oviducts. Epithelial cells isolated from oviducts collected during the follicular phase were cultured in M199 under standard culture conditions until monolayer formation. The cells were trypsinized and plated at a density of 3 X104 /ml/well and cultured again until subconfluency, at which time the cells were incubated for 4 or 24 h with M199 only (control), high progesterone (1μg/ml), low progesterone (10 ng/ml), oestradiol-17beta (1 ng/ml), LH (10 ng/ml), oxytocin (10-9 M) endothelin-1 (10-9 M), PGE2 (10-8 M) PGF2alpha (10-9 M) or their combination (high progesterone + oestradiol-17beta, low progesterone + oestradiol-17beta, LH + oestradiol-17beta, endothelin-1 + oestradiol-17beta, low progesterone + oestradiol-17beta + LH and high progesterone + oestradiol-17beta + LH). The production ofPG and endothelin-1 was increased by oestradiol-17beta + low progesterone and LH + oestradiol-17beta + low progesterone (P<0.05); LH + oestradiol-17beta increased production of PGF2alpha and endothelin-1 (P<0.05). Oestradiol-17beta + endothelin-1 stimulated PG production (P<0.05). Oxytocin had no effect on the production of any of these substances. It is suggested that the preovulatory LH surge, together with locally re-circulated high levels of oestradiol-17beta from Graafian follicles and basal progesterone from regressing corpus luteum, induces the maximum stimulatory effect on oviductal PGE2, PGF2alpha and endothelin-1 production during the periovulatory period and that the elevated local endothelin-1 concentration during the periovulatory periodinduces the high contractile activity of the oviduct and, at the same time, the stimulation of PG production.
