高氧环境下肺成纤维细胞RB蛋白磷酸化状态的改变

         

摘要

目的:探讨新生大鼠在高氧环境下肺成纤维细胞(LF)RB蛋白磷酸化状态的改变.方法:足月新生SD大鼠于生后12h内随机分为空气对照组和高氧组,分别持续吸入空气和体积分数为0.85的氧气,于3、7、14、21 d随机处死动物后,无菌条件下采集肺组织,进行LF的原代培养,应用免疫组化分别检测总RB蛋自及磷酸化RB蛋白(p-RBser-795)表达.结果:高氧组与空气组相比,各时点的LF总RB蛋白表达差异无统计学意义(P>0.05),p-RB于生后3d空气组与高氧组比较差异无统计学意义,生后7~21 d时高氧组表达开始增多,与对照组比较差异有统计学意义(P<0.05或P<0.01).结论:高氧可能导致RB蛋自磷酸化(RB灭活),LF过度增殖最终导致肺间质纤维化.%Objective: To explore the variation of retinoblastoma (RB) phosphorylation in lung fibroblasts (LF) of neonatal rats treated with hyperoxia. Methods: About 12 hours after birth, full-term newborn rats were randomly divided in two groups, hyperoxia group and room air group, and continuously exposed to oxygen (0.85) or room air. Rats were randomly selected to be sacrificed on postnatal 3 d, 7 d, 14 d and 21 d. The lung tissues were immediately resected under sterile condition,and primary cells were cultured. The protein expressions of RB and phosphorylated RB (p-RB, ser-795) were detected by immunohistochemistry. Results: The levels of RB in LF at different time points were no differences hetween hyperoxia group and room air group (P > 0.05). And there were no significant difference in expression of p-RB on postnatal 3 d between hyperoxia group and room air group. The expression of p-RB was higher gradually 7-21 d after birth in hyperoxia group and showed a significant difference compared with that of control (P < 0.05 or P < 0.01). Conclusion: The phosphorylation of RB protein can be induced by hyperoxia. Excessive prolif'eration of LF leads the pulmonary interstitial fibrosis.
