胰岛破坏大鼠模型的血糖变化

         

摘要

目的 探讨胰岛破坏与血糖的关系.方法 将46只雄性SD大鼠随机分为实验组36只和对照组10只,实验组按60 mg/kg一次性腹腔内注射链脲佐菌素,对照组注射等量的缓冲液;在注射后的第7、14、28、42、56、84天检测空腹血糖并行腹腔葡萄糖耐量试验(IPGTT);在第84天同时检测血清胰岛素,并取胰腺制作树脂切片;光镜下定性观察胰岛形态,用体视学方法测量胰岛的大小及总体积等.结果 实验组成模大鼠19只为高血糖组、未成模大鼠10只又分为血糖正常组和血糖部分正常组各5只.与对照组比较,高血糖组大鼠(14.9 mmol/L≤血糖≤27.8 mmol/L)的胰岛平均面积、单位面积胰腺内胰岛数量和胰岛总体积分别减少了64.3%、68.2%和91.3% (P均<0.05),血清胰岛素减少了92.6%;血糖部分正常组大鼠(3.6 mmol/L≤血糖≤27.8 mmol/L)的IPGTT均异常,第56、84天的血糖分别升高了247.2%和313.2%(P均<0.05),血清胰岛素、胰岛总体积分别减少了94.0%、86.4%(P均<0.05);血糖正常组大鼠(4.5 mmol/L≤血糖≤7.9 mmol/L)的血糖、血清胰岛素和胰岛总体积无明显变化,但部分IPGTT异常.结论 血糖水平与胰岛破坏的程度有关.%Objective To explore the correlation of the damaged pancreas islet and the level of blood glucose. Methods Forty-six ordinary male SD rats were randomly divided into control group and experimental group. Rats in the experimental group received intraperitoneal injection of 60 mg/kg STZ and rats in the control group received intraperitoneal injection of equal dose acidified saline. At day 7, 14, 28, 42, 56 and 84 after injection, observation on fasting blood glucose and IPGTT were performed. At day 84, serum insulin was detected, the pancreata were removed after excision and hydroxyethyl methacrylate resin-embeded pancreas sections were obtained. The histology of pancreas islet was observed by light microscopy. The area and total volume of the pancreas islet were measured using stereological methods. Results Compared with the control group, average areas of islet, numbers of islet of unit of area in pancreas and the total volumes of pancreas islet were respectively decreased 64. 3% , 68. 2% and 91. 3% (P <0.05) and serum insulin levels were decreased 92.6% in hyperglycemic rats (14. 9 mmol/L ≤ blood glucose levels ≤27. 8 mmol/L) in the experimental group. Compared with the control group, rats with abnormal blood glucose levels (3. 6 mmol/L ≤ blood glucose levels≤27. 8 mmol/L) had abnormal IPGTT. Their blood glucose levels at day 56 and 84 were respectively increased 247. 2% and 313.2% (P< 0.05) and their serum insulin levels and the total volumes of pancreas islet were respectively decreased 94.0% and 86.4% (P <0.05). There were no statistical differences in the blood glucose, serum insulin levels and the total volumes of pancreas islet between the control and the rats with normal blood glucose levels (4.5 mmol/L≤ blood glucose levels≤7.9 mmol/L), but some of them had abnormal IPGTT. Conclusion There is positive relativity between damaged degree of pancreas islet and blood glucose level.
