目的:观察茶多酚对人喉鳞癌细胞株Hep-2增殖、凋亡的影响,并探讨其作用机制。方法培养Hep-2细胞,加入不同浓度的茶多酚共育,比较细胞增殖抑制率、细胞核形态,并比较细胞内Caspase-3、Bax、Bcl-2 mRNA的相对表达量。结果茶多酚可抑制Hep-2细胞的增殖,诱导Hep-2细胞凋亡,上调Caspase-3、Bax mRNA表达,下调Bcl-2 mRNA表达,且呈浓度依赖性(P均<0.05)。结论茶多酚能抑制Hep-2细胞增殖,并诱导其凋亡,其机制可能与Caspase-3、Bax mRNA表达上调和Bcl-2 mRNA表达下调有关。%Objective To observe the effects of tea polyphenol ( TP) on the proliferation and apoptosis of human la-ryngeal cancer Hep-2 cells, and to investigate its mechanism. Methods The Hep-2 cells were cultivated, and were treated with different concentrations of TP. The proliferation inhibition rate and the nuclear shape of Hep-2 cells were compared, and the relative expression of Caspase-3, Bax and Bcl-2 mRNA in cells was compared. Results TP inhibited the prolifera-tion, induced apoptosis of Hep-2 cells, up-regualted the expression of Caspase-3 and Bax mRNA, and down-regulated the expression of Bcl-2 mRNA, which was in a dose-dependent manner. (all P<0. 05). Conclusions TP can inhibit the proliferation of Hep-2 cells, and induce its apoptosis. The mechanism may be related to the up-regulated expression of Caspase-3 and Bax mRNA and down-regulated expression of Bcl-2 mRNA.
