木蝴蝶总黄酮的提取及体外清除亚硝酸盐及阻断亚硝胺合成的研究

             

摘要

目的:优化木蝴蝶中总黄酮的提取工艺,研究木蝴蝶总黄酮、水提液、精油,清除亚硝酸盐的效果及阻断亚硝胺合成的能力。方法在单因素实验的基础上,以超声时间、料液比和乙醇体积分数为自变量,以总黄酮提取率为因变量,对自变量各水平进行多元线性回归和二项式拟合,通过响应面曲线法选择较佳工艺;利用紫外分光光度法检测研究木蝴蝶总黄酮、水提液、精油对清除亚硝酸盐及阻断亚硝胺合成的影响。结果木蝴蝶总黄酮的较佳提取条件为:料液比1∶53,超声时间119 min ,乙醇体积分数45%,木蝴蝶中总黄酮、精油及水提物对亚硝酸钠的最大清除率分别为65.3%,71.2%和42.2%,对亚硝胺合成的最大阻断率分别为58.5%,36.7%和30.9%。结论木蝴蝶总黄酮对亚硝酸盐的最大清除率略低于精油,但高于大蒜,达到65.3%。同时抑制亚硝化反应的效果高达58.5%,高于精油,其阻断率接近大蒜,清除率和阻断率都高于水提液。木蝴蝶中总黄酮具有较好的清除能力和阻断效果。%Objective To optimize the extraction process of total flavonoids in Oroxylum indicum ,and to study the effect of total flavonoids from Oroxylum indicum on the nitrite savenging and synthesis blocking of nitrosamine .Methods On the basis of single factor ,the re‐sponse surface method was used .UV spectrophotometry was used to study the effect of the water extract and essential oil on the nitrite scavenging and nitrosamine synthesis blocking .Results The optimal extraction conditions of total flavonoids from Oroxylum indicum were as follows :the ratio of material to liquid 1∶53 ,ultrasonic time 119 min ,ethanol concentration 45% .The maximum scavenging rate of sodium nitrite were 65 .3% ,71 .2% and 42 .2% respectively ,the maximum blocking synthesis of nitrosamine was 58 .5% ,36 .7% and 30 .9% respectively .Conclusion The effect of flavonoids on nitrite scavenging was slightly lower than that of essential oil ,but higher than that of garlic ,reaching up to 65 .3% .At the same time ,the effect of inhibition of nitrosation was up to 58 .5% ,higher than that of oil .The blocking rate closed to the garlic .The clearance rate and blocking rate was higher than the water extract .The flavonoids have good effect on scavenging and blocking .
