高效液相指纹图谱结合化学计量学评价不同产地芡实的质量

         

摘要

Objective To establish a method for the quality evaluation of Euryale semen from different areas by fingerprint combined with chemometrics.Methods 33 samples were collected from 11 provinces.HPLC chromatograms were obtained.The common pattern was established for chemometrics analysis.Similarity analysis,2D hierarchical cluster analysis, principal component analysis were performed.Results Euryale semen from 11 provinces could be classified by common pattern which contained 9 characteristic peaks using similarity analysis.The similarity of Shandong,Hunan,Jiangxi,Anhui and Hebei provinces were over 0.90,while the similarity of other areas were less than 0.90.It showed the significant differences between samples.No.1,3,4 peaks were the most important peaks which could be used to identify the differences between areas by hot maps of hierarchical cluster analysis and loading chart of principal component analysis.Samples from Jiangsu province were more stable than from other areas.Conclusion This method is flexible and comprehensive,and could differentiate and evaluate Euryale semen of different areas without reference standard.It could be used to control the quality of Euryale semen.%目的 建立指纹图谱结合化学计量学评价不同产地芡实的质量.方法 收集全国11个省33批芡实,建立统一条件的液相指纹图谱和部分特征峰的共有模式,计算相似度,进行二维聚类分析和主成分分析来寻找导致产地差异的特征峰.结果 由9个特征峰组成的共有模式进行相似度评价可将11个省的芡实分开,其中山东、湖南、江西、安徽和河北的样本相似度均高于0.90,而其他地域的样本均小于0.90,体现出不同地域的芡实差异较大;由二维聚类得到的热图和主成分分析得到的载荷图可发现,1,3,4号峰是有效体现不同地域芡实样本差异的强特征峰.其中江苏样本较为稳定,而其他地方样本离散度较大.结论 该多元统计分析方法较灵活、全面,无需对照品即可区分和评价不同产地的芡实,可为该类药材的质量控制提供参考.
