人胎盘源性间充质干细胞体外分离培养及多向分化潜能的研究

     

摘要

To study the biological character of mesenchymal stem cells derived from human placenta. The mesenchymal stem cells were isolated from human placenta by cell adherence. The cell cycle and surface antigens were detected by flow cytometry. The cells were induced to differentiation adipocytes, osteocytes and nerve cells. The mesenchymal stem cells could be cultured and proliferated in vitro by cells adherence. Flow cytometry analysis revealed that the cells expressed CD29 and CD44, while CD34, CD133.HLA-DR and vWF were negative. Nearly 77% of the mesenchymal stem cells were in G0/G1 phase. After induction,oil red O staining and alizarin red S staining were positive,and the nerve cells expressed NSE, respectively. Human placenta is a-bundant with mesenchymal stem cells,which can be isolated and the biological character is similar to other source of mesenchymal stem cells.%为体外分离培养人胎盘源性的间充质干细胞,并对其生物学特性进行研究.采用组织块贴壁培养法分离培养获取胎盘源性间充质干细胞,通过培养扩增后用流式细胞仪检测细胞周期及免疫表型,在体外诱导其向成骨细胞、脂肪细胞、神经细胞方向分化,并通过特异性染色鉴定其分化能力.结果显示采用组织块贴壁法能成功获得贴壁生长的成纤维样人胎盘源性间充质干细胞;流式细胞本分析结果示,人胎盘源性间充质干细胞高表达CD29、CD44,阴性表达CD34、CD133、HLA-DR及vWF,细胞周期结果示,G0/G1期细胞约占77%;经向脂肪细胞诱导后油红O染色阳性,向成骨细胞诱导后茜素红染色为阳性,向神经细胞诱导后,NSE表达阳性.由此可知,人胎盘中富含间充质干细胞,与其他来源的间充质干细胞的生物学特征相似.
