脱钙骨关节诱导关节软骨再生的蛋白质组学初步研究

         

摘要

Objective To investigate the mechanism of the reproduction of articular cartilage by verifying the specific proteins emerging in the process of demineralized osteoarticular and articular cartilage transplantation. Methods 24 New Zealand white rabbits were divided into 3 groups, the defects and demineralized osteoarticular and articular cartilage transplantations were administered inⅠ and Ⅱ group, which were served as experimental groups, the other was the control group. The synovia of the 3 groups were collected in 8th, 12th and 16th week, and were assessed by 2-D electrophoresis and mass chromatographic analysis. Results As compared to that in control group, two unnamed differential proteins whose molecular mass were 15 313.8 and 11 337.7 were found in the experimental groups. Conclusion In the process of demineralized osteoarticular and articular cartilage transplantation, 2 differential proteins which may play important roles in the reproduction of articular cartilage were found.%目的:通过分析在脱钙骨关节及软骨诱导关节软骨再生的过程中宿主所产生的特异性蛋白,探讨关节软骨再生的机制.方法:将24只新西兰大白兔分为3组,Ⅰ、Ⅱ组为实验组,分别对其关节软骨面进行破坏,并在关节腔内植入脱钙骨关节或软骨,Ⅲ组为空白对照组,分别于第8、第12、第16周抽取各组关节液,进行双向电泳及质谱分析.结果:与空白对照组相比,两个实验组中分别出现了分子量为15313.8与11 337.7的两种未命名的差异蛋白.结论:在植入脱钙骨关节以及软骨的宿主关节滑液中产生了与关节软骨再生可能密切相关的差异蛋白,为进一步研究奠定了基础.
