RP-HPLC法同时测定小儿肺热咳喘颗粒中两种主要活性成分的含量

         

摘要

目的:建立HPLC法测定小儿肺热咳喘颗粒中2种活性成分的含量。方法色谱柱为PLATSILTM ODS色谱柱(250 mm ×4.6 mm,5μm),柱温30℃,流动相为甲醇-0.1%磷酸水溶液,采用梯度洗脱:0~5 min,35%~50%A;5~10 min,50%~75%A;10~17 min,75%~90%A;17~25 min,90%A;25~30 min,90%~35%A;30~40 min,35%A,检测波长为327 nm,流速为1 mL/min,柱温30℃。结果绿原酸、黄芩苷的线性范围分别为0.084~1.260μg (R=0.9995)和0.242~3.630μg(R=0.9996),此范围内与峰面积呈良好的线性关系。结论该法检测效率高,专属性强,重复性好,可用于小儿肺热咳喘颗粒中绿原酸和黄芩苷的定量测定。%Objective To establish a method for simultaneous determination of chlorogenic acid and baicalin in Xiao'er Feire Kechuan Granules .Methods Using PLATSILTM ODS (250 mm ×4.6 mm,5μm) column with metha-nol (A) -0.1%phosphoric acid (B) gradient:0 ~5 min,35%~50%A;5~10 min,50%~75%A;10~17 min, 75%~90%A;17~25 min,90%A;25~30 min,90%~35%A;30~40 min,35%A,a flow rate of 1 mL/min,detec-tion wavelength:327 nm, the column temperature was 30 ℃.Results The results showed that the linear range of chlorogenic acid and baicalin was 0.084~1.260μg (R=0.999 5) and 0.242~3.630μg (R=0.9996) respec-tively ,and the sample amout and the peak area had a good linear relationship .Conclusion The method is accurate , simple,with good reproducibility and stability ,and is suitable for quality control of Xiao'er Feire Kechuan Granules .
