Rapid and sensitive reverse phase high performance liquid chromatography(RP-HPLC) method with UV detection for quantification of four components indihydroergotoxine mesilate for injections have been developed and validated.Chromatography was performed using COSMOSIL C18 (25 cm x 4.6 mm i.d.5 pm particle size) column as a stationary phase and water-acetonitrile-triethylamine (150:50:5, v/v) as a mobile phase, maintaining the flow rate at1.5 mL min~-1 with UV detection at 280 nm. The proposed method wassuccessfully applied for the determination of dihydroergotoxine mesilate in theirinjections form over a concentration range of 120.17-480.67 ug mL~-1 with meanpercentage recovery of over 99%. The RSD was below 1.5% (n = 9). The RP-HPLC method was simple, highly sensitive, precise, and accurate, suggesting thatthe developed method is useful for routine quality control.
