小檗碱衍生物B-119抑制肿瘤及血管内皮细胞增殖的作用

         

摘要

Objective To investigate the effect of berberine derivate B-119 against the proliferation of tumor cells and against the proliferation, migration and tube-like structure formation of vascular en-dothelial cells. Methods MTT assay was applied to screen the anti-proliferative effect of B-119 on 8 kinds of tumor cell and ECV304 cells in vitro. The impact of B-119 on the migration of ECV304 cells was measured by scarification test, and the impact of B-119 on tube-like structure formation of ECV304 cells was examined under microscope. Results B-119 had different degrees of inhibition to these 8 tumor cell lines, and the IC50 of B-119 to these tumor cells were 0. 76~26. 83 mg/L. B-119 could inhibit the proliferation of ECV304 cells in a concentration-dependent and time-dependent manner. After being treated with B-119 for 24 h, 48 h and 72 h, MTT assay showed the IC50 of B-119 to ECV304 were 59. 22, 12. 08 and 3. 31 mg/L respectively. The migration of ECV304 was reduced, and the inhibition effect of B-119 on ECV304 cells was strengthened with the increase of concentrations. After being treated with B-119 at the concentrations of 1. 25 , 2. 5 and 5 mg/L for 24 h, the number of ECV304 cells migration was gradually decreased and the migration inhibition ratios were 49. 29%, 59. 71% and 71. 94% respectively (P<0. 01). B-119 could inhibit the tube formation of ECV304 cells, After being treated with B-119 at the concentrations of 10, 20 and 30 mg/L for 18 h, the tube formation inhibition ratios were 35. 71%, 57. 14% and 67. 86% respectively (P<0. 01). Conclusions B-119 could inhibit the proliferation of 8 kinds of tumor cell and also inhibit the proliferation, migration and tube-like structure formation of ECV304 cells.%目的 探讨小檗碱衍生物B-119对肿瘤细胞的增殖抑制作用及对血管内皮细胞增殖、迁移和小管形成的影响.方法 MTT法检测B-119对瘤细胞株MDA-MB-231、B16、K562、LLC、HT29、SMMC-7721、Colo205、HL60和血管内皮细胞ECV304的增殖抑制作用;划痕法测定B-119对血管内皮细胞ECV304迁移的影响;观察B-119对血管内皮细胞ECV304小管形成的影响.结果 B-119对8株肿瘤细胞具有不同程度的抑制作用,IC50值为0.76~26.83 mg/L;B-119对ECV304的增殖抑制作用表现为时间、浓度依赖性,24,48,72 h的IC50分别为59.22,12.08,3.31 mg/L;药物干预组ECV304细胞迁移数减少,且随B-119浓度的增加,迁移抑制作用越强,B-119浓度为1.25,2.5,5 mg/L干预24 h后,细胞迁移数渐次减少,抑制率分别为49.29%,59.71%和71.94%(P<0.01);B-119对ECV304小管形成有明显的抑制作用,浓度为10,20,30 mg/L时的小管形成抑制率分别为35.71%,57.14%和67.86%(P<0.01).结论 B-119可抑制肿瘤细胞的增殖,并抑制血管内皮细胞的增殖、迁移和小管形成.
