响应面法优化黑曲霉固态发酵产木聚糖酶工艺

         

摘要

The xylanase production with Aspergillus niger FIP-09-24 in solid fermentation with wheat bran and barley as substance.The single factor experiments test and Plackett-Burman test suggested that carbon,nitrogen and water were the major factors on the synthesis of xylanase.Three-level response surface methodology based on the principle of central composite design was employed to analyze the significance and interactions of the above factors by mathematical modeling.The optimal conditions were determined by response surface analysis,which was the mass ratio of 3.8 ∶ 1 between wheat bran and barley residue,55.7% of water content,2.0% of nitrogen content,temperature 28 ℃,incubation time 60 h.The enzyme activity of the fermentation culture can reach up to 66 002 U/g.%采用固体发酵法探索了黑曲霉(Aspergillus niger FIP-09-24)作用于基质麸皮和大麦渣生产木聚糖酶的最佳工艺条件.通过单因素试验和Plackett-Burman试验确定了碳源、含水量和氮源3个主要因素对固体发酵合成木聚糖酶的影响,根据中心组合设计原理采用三因素三水平的响应面分析法,获得了黑曲霉固体发酵产木聚糖酶的最佳工艺条件.结果表明,麸皮和大麦渣质量比为3.8∶1、含水质量分数55.7%、含氮质量分数2.0%、28℃培养60 h,发酵曲的木聚糖酶活力最高,为66 002 U/g.
