茵陈蒿汤血清药化学研究∗

     

摘要

Objective To analyze the constituents of Yinchenhao Decoction ( YCHD ) absorbed into blood by serum pharmacochemistry. Methods SD rats were used as donor and serum containing drugs were prepared after intragastric administration of YCHD, YCHD without one of the component herbs, and individual herbs of YCHD. HPLC fingerprints of serum containing drug were established. By comparing the fingerprints of serum containing YCHD with the one of control serum, drug-induced constituents in blood were distinguished. By comparing the fingerprint of serum containing YCHD with the ones of serum containing YCHD without either one of the component herbs, and individual herbs of YCHD, the sources of drug-induced constituents were analyzed. By comparing the retention time and spectrograms of chroma-tographic peaks in fingerprint of serum containing YCHD with the ones of YCHD and reference sub-stances, drug-induced constituents in blood were identified. Results 23 drug-induced constituents in serum containing YCHD were detected. 6 of them were original compounds, other 17 were metabolites. 3 pounds were identified as anthraquinone from Rhei Radix et Rhizoma( RRR) and 2 crocetin derivatives from Gardeniae Fructus( GF) . Out of 17 metabolites, 1 was identified as the metabolite of iridoid, 7 were identified as the metabolites of anthraquinone, and 9 were identified as the metabolites of crocetin deriva-tives. Conclusion Drug-induced constituents in serum containing YCHD include original form of an-thraquinones from RRR, original form of iridoids and crocetin derivatives from GF, and the metabolites of the constituents mentioned above. All these drug-induced constituents may be the pharmacodynamic ma-terial basis for hepatoprotective effects of YCHD.%目的:采用血清药化学方法对茵陈蒿汤( YCHD)的入血成分进行分析。方法以SD大鼠为供体,灌胃给药制备YCHD及各缺味方、单味药含药血清,建立含药血清HPLC指纹图谱。与空白血清指纹图谱比对,指认含药血清中的药源性成分;与各缺味方、单味药含药血清比对,分析各药源性成分来源;与体外指纹图谱、对照品指纹图谱色谱峰保留时间及紫外吸收光谱图指纹图谱比对,对各药源性成分进行鉴别。结果 YCHD含药血清中检测到23个药源性成分,其中6个为体外原形成分,17个为体内代谢产物。6个原形成分分别为来源于栀子的京尼平苷酸、栀子苷、西红花酸类成分,来源于大黄的大黄酸及蒽醌类成分。17个代谢产物中的1个为环烯醚萜类成分代谢产物,7个为蒽醌类成分代谢产物,9个为西红花酸类成分代谢产物。结论 YCHD含药血清中的药源性成分主要为来源于大黄的蒽醌类成分、来源于栀子的环烯醚萜类成分和西红花酸类成分及以上成分的代谢产物,这些药源性成分可能构成了该方保肝作用的药效物质基础。
