高效液相色谱法测定牛黄解毒滴丸中大黄素和大黄酚含量

摘要

Objective To establish a HPLC method for determination of emodin and chrysophanol in niuhuang jiedu dropping pill.Methods Kromasil C18( 4.6 mm×l50 mm,5 μm ) column was used; the mobile phase was methanol-0.1％ phosphoric acid ( 85: 15 ); the flow rate was 1 mL · min-1 and column temperature was at 30 C; the detection wave-length was 254 nm.Results There was a good linear relationship of emodin and chrysophanol at range from 2.36-37.72 μg · mL-1 and 6.41-102.60 μg · mL-1 , respectively; the average recoveries were 99.56％ ( RSD= 1.1％ , n= 9 )and 99.89％ ( RSD= 1.8％ ,n =6 ), respectively.Conclusion This method is convenient, accurate and reproducible for determination of emodin and chrysophanol in niuhuang jiedu dropping pill.%目的 采用高效液相色谱法测定牛黄解毒滴丸中大黄素和大黄酚含量.方法 色谱柱:Kromasil C18柱(4.6 mm×150 mm,5 μm);流动相:甲醇-0.1%磷酸溶液(85:15);流速:1 mL·min-1;紫外检测波长:254 nm;柱温:30 ℃.结果 大黄素在2.36～37.72 μg·mL-1范围内线性关系良好,平均回收率分别为99.56%,RSD=1.1%(n＝9);大黄酚在6.41～102.60 μg·mL-1范围内线性关系良好,平均回收率99.89%,RSD=1.8%(n＝9).结论 该方法简便,准确,可作为牛黄解毒滴丸质量控制的方法.
